摘 要:为分离更多高效的锑氧化菌,本研究采用传统微生物分离的方法从锑矿区土壤中分离了8株耐锑微生物,分别命名ZH1~ZH8.且利用氧化性鉴定发现5株菌具有较强的锑氧化能力.对分离的8株菌株进行了基于分子生物学的菌种鉴定,并分析了5株锑氧化菌的氧化效率以及生长特性.结果发现,分离的8株菌分别属于假单胞菌属(Pseudomonas),不动杆菌属(Acinetobacter)和贪铜菌属(Cupriavidus).氧化效率测定结果发现,5株锑氧化菌中,ZH4的氧化性最强,在培养的第十天将培养基的Sb(Ш)氧化95%.正交试验结果显示,ZH4的最佳生长条件为:温度28℃,pH5.2,锑浓度50μmol/L,盐度1%.In order to separate more Sb(Ⅲ)-oxidizing bacteria,8 Sb(Ⅲ)-resistant bacteria were isolated from the antimony contaminated soil by traditional microbial separation method,which was named as strains ZH1~ZH8.Five strains with strong oxidative capacity were found out by oxidative identification.The phylogenetic study of 8 Sb(Ⅲ)-resistant bacteria were analyzed.The results showed that 8 strains belonged to Pseudomonas,Acinetobacter,and Cupriavidus respectively.The oxidation efficiency and growth characteristics of 5 Sb(Ⅲ)-oxidizing bacteria were identified.Among 5 strains,ZH4 strain had the strongest oxidation ability,it was able to oxidize 95% of the Sb(Ⅲ) to Sb(Ⅴ) on the tenth day of culture.The optimal growth conditions of the most oxidized ZH4 bacteria were determined by orthogonal test.The results showed that ZH4 bacteria grew optimally at a temperature of 28 ℃,a pH of 5.2,an antimony concentration of 50 μ mol/L,and a salinity of 1%.
