摘 要:目的研究雷公藤多苷片诱导卵巢早衰模型小鼠的自身修复性变化,为深入研究POF的病理生理机制和预防措施提供实验依据。方法筛选36只动情周期正常的Balb/c小鼠,随机分为阴性对照组、模型组。模型组小鼠按50 mg/kg灌胃给予的雷公藤多苷片溶液,1次/d,连续14天。于造模14 d,停止造模27 d和35 d,每组随机取6只小鼠,取血检测FSH、LH、E2含量,取双侧卵巢,测量脏器系数,病理检查卵巢的组织形态学变化。结果造模期间小鼠均表现为行动迟缓、精神萎靡,脱毛,倦怠等,体质量减轻,性周期紊乱率100%。停止造模15 d后,动物状态、性周期逐步恢复正常。与阴性对照组比较,造模14 d、停止造模27 d、停止造模35 d,小鼠血清中E2水平、卵巢系数降低差异有统计学意义(P<0.01);小鼠血清中FSH水平和LH水平升高差异有统计学意义(P<0.01)。在造模14 d,小鼠卵巢组织萎缩,皮质髓质结构混乱,各级卵泡数量减少,闭锁卵泡增多、卵母细胞及透明带形态异常;黄体数量减少,形态不规律。停止造模28 d和35 d,模型组小鼠除黄体数量正常外,其余均未见明显变化。结论雷公藤多苷片造模14 d可导致小鼠卵巢早衰,停止造模15 d后状态逐步恢复正常,但激素水平以及卵巢脏器至停在造模35 d尚未恢复正常。Objective To study the recovery of premature ovarian failure mice model induced by Tripterygium wilfordiipolyglycosides.Methods 36 Balb/c mice with regular oestrous cycle were randomly into control group and model group. Model group was intragastric administration by Tripterygium wilfordiipolyglycosides (50mg/kg) once per day for 14 days. 6 mice of each group were sacrificed to collect the serum and ovary on the day when the administration ended and the 27th and 35th day after the last administration.The FSH, LH and E2 concentration in serum were measured. And the histological structure of ovary was observed.Results During the administration, drumble, spirits atrophy, hair lost and weight lost could be observed in model group. Also the oestrous cycle of model group were disordered. 15 days after the administration, the mice of model group recovered gradually. Compared with control group, serum-E2 and ovarian index of model group decreased while serum-FSH and LH increased ( P <0.01). After administration for 14 days, ovarian atrophy,follicle decreasing, atretic follicle increasing, zona pellucid anomaly and corpus luteum decreasing were observed in model group. The 28th and 35th day after the last administration, the amount of corpus luteum in model group recovered but the other changes were retained.Conclusion The administration of Tripterygium wilfordiipolyglycosides on mice for 14 days can induced the premature ovarian failure. 15 days after the administration the mice of model group recovered gradually but the hormone level and ovarian structure did not get back to normal till the 30th day.
