摘  要:目的观察5-羟色胺(5-HT)对肺动脉平滑肌细胞瞬时2受体电位通道(TRPC)/活化T细胞核因子(NFAT)蛋白表达的影响。方法取肺动脉平滑肌细胞,分别采用不同浓度(0μmol/L、0.1μmol/L、1μmol/L、10μmol/L、33μmol/L、100μmol/L)5-HT、不同浓度(0μmol/L、0.01μmol/L、0.1μmol/L、1μmol/L、3.3μmol/L、10μmol/L)的5-HT2A受体拮抗剂酮色林+10μmol/L5-HT和不同浓度(0μmol/L、0.1μmol/L、1μmol/L、10μmol/L、33μmol/L、100μmol/L)的TRPC抑制剂SKF96365+10μmol/L5-HT进行处理。24h后,检测各组细胞中TRPC1、TRPC6和NFATc3的蛋白表达水平。结果与0μmol/L5-HT相比,10μmol/L、33μmol/L、100μmol/L5-HT干预后细胞中TRPC1、TRPC6、NFATc3蛋白表达水平均升高(均P<0.05)。与0μmol/L酮色林+10μmol/L5-HT比较,1μmol/L、3.3μmol/L、10μmol/L酮色林+10μmol/L5-HT干预后细胞中TRPC6及NFATc3蛋白表达水平均降低(均P<0.05)。与0μmol/LSKF96365+10μmol/L5-HT比较,10μmol/L、33μmol/L、100μmol/LSKF96365+10μmol/L5-HT干预后TRPC6、NFATc3蛋白表达水平均降低(均P<0.05)。结论10~100μmol/L的5-HT均可上调肺动脉平滑肌细胞TRPC1、TRPC6和NFATc3蛋白的表达水平,且应用一定浓度的5-HT2A受体拮抗剂酮色林或TRPC抑制剂SKF96365均可抑制5-HT对其中TRPC6和NFATc3蛋白的表达。Objective To observe the effects of 5-hydroxytryptamine(5-HT) on the protein expression of transient receptor potential channel(TRPC)/nuclear factor of activated T cell(NFAT) in pulmonary artery smooth muscle cells(PASMCs).Methods PASMCs were obtained and treated with 5-HT of different concentrations( 0 μmol/L,0.1 μmol/L,1 μmol/L,10 μmol/L,33 μmol/L and 100 μmol/L),10 μmol/L 5-HT plus 5-HT 2A receptor antagonist ketanserin of different concentrations(0 μmol/L,0.01 μmol/L,0.1 μmol/L,1 μmol/L,3.3 μmol/L and 10 μmol/L),and 10 μmol/L 5-HT plus TRPC inhibitor SKF96365 of different concentrations(0 μmol/L,0.1 μmol/L,1 μmol/L,10 μmol/L,33 μmol/L and 100 μmol/L).After 24 hours,the expression levels of TRPC1,TRPC6 and NFATc3 proteins were detected in the PASMCs of each group.Results Compared with 0 μmol/L 5-HT,intervention with 10 μmol/L,33 μmol/L or 100 μmol/L 5-HT resulted in elevated expression levels of TRPC1,TRPC6 and NFATc3 proteins in PASMCs(all P <0.05).Compared with 0 μmol/L ketanserin plus 10 μmol/L 5-HT,intervention with 10 μmol/L 5-HT plus 1 μmol/L,3.3 μmol/L or 10 μmol/L ketanserin resulted in decreased expression levels of TRPC6 and NFATc3 proteins in PASMCs(all P <0.05).Compared with 0 μmol/L SKF96365+10 μmol/L 5-HT,intervention with 10 μmol/L 5-HT plus 10 μmol/L,33 μmol/L or 100 μmol/L SKF96365 resulted in decreased expression levels of TRPC6 and NFATc3 proteins in PASMCs(all P <0.05).Conclusion 5-HT with the concentration of 10-100 μmol/L can up-regulate the expression levels of TRPC1,TRPC6 and NFATc3 proteins in PASMCs,and administration of 5-HT 2A receptor antagonist ketanserin or TRPC inhibitor SKF96365 with a certain concentration can inhibit the effect of 5-HT on the expression of TRPC6 and NFATc3 proteins in PASMCs.
