摘 要:目的探讨华蟾毒精对人结直肠癌SW-480、HT-29、HCT-116细胞的放疗增敏作用.方法选用SW-480、HT-29、HCT-116这3种人结直肠癌细胞株,细胞经单纯放疗和联合药物处理24 h、48 h、72 h后CCK8法检测细胞增殖,并于处理后48 h流式细胞仪测定细胞周期和凋亡.结果与单独放疗作用相比,华蟾毒精和放疗联合应用能明显抑制人结直肠癌SW-480、HT-29、HCT-116细胞增殖,同时诱导和促进人结直肠癌细胞的凋亡发生,调整细胞周期的变化.结论华蟾毒精可以增加人结直肠癌SW-480、HT-29、HCT-116细胞的放疗敏感性,提高放疗对人结直肠细胞的放射治疗效果.Objective To assess the effects of cinobufagin on enhancing the radiosensitivity of human colorectal cancer cells in vitro. Methods SW-480,HT-29 and HCT-116 cells were exposed to X-ray with or without cinobufagin treatment respectively. CCK8 assay was used to evaluate the cell viability. And flow cytometry was performed to assess the cell cycle and apoptosis. Results Compared with the exposed cells without cinobufagin, the proliferationof SW-480,HT-29 and HCT-116 cells treated with cinobufagin weresuppressedsignificantlywith cell apoptosis being induced,which resulted in the change of cell cycle. Conclusions Cinobufagin can enhance the radiosensitivity of SW-480,HT-29 and HCT-116 cells which increases the efficiency of radiotherapy with X-ray irradiation.
