摘  要:将清洗消毒后金花茶种子,经保留与不保留种皮的两种处理,培养于4种介质中,筛选最佳无菌萌发方案;以无菌苗叶片诱导愈伤组织,采用WPM基础培养基添加不同激素,筛选出最佳的愈伤组织诱导培养基配方;比较MS和WPM培养基,筛选丛生芽增殖培养优化培养基。结果显示,将去除种皮的种子接种于无菌水+珍珠岩的培养介质中进行无菌萌发效果最好,萌发率可达80%;最佳的叶片愈伤组织诱导培养基为WPM+NAA0.1mg/L+2,4-D0.2mg/L;最佳的丛生芽增殖培养基为WPM+6-BA1mg/L+NAA0.05mg/L。The cleaned and disinfected camellia nitidissima seeds were cultured in 4 media with or without seed coat,and the best aseptic germination scheme was selected;Aseptic seedling leaves was used as explant,and WPM basal medium was added with different hormones to select the best formula of callus induction medium.The MS and WPM media were compared to screen the optimal media for cluster bud proliferation culture.The results are as follows:Seed with seed coat removed was inoculated in the culture medium of sterile water+perlite for sterile germination with the best effect.The germination rate reached 80%;The best medium for callus induction with leaf was WPM+NAA 0.1 mg/L+2,4-D 0.2 mg/L;the best medium for proliferation of cluster buds was WPM+6-BA 1 mg/L+NAA 0.05 mg/L.
