摘 要:细胞外基质(ECM)作为一种特殊的天然生物衍生材料,为细胞提供了生物物理机械性支持,并且可为组织的再生提供良好的微环境.通过使用Ⅰ型胶原酶以及植入动物实验对脱细胞猪皮基质材料进行体内外降解研究,分析脱细胞猪皮基质材料的降解规律以及力学性能.结果显示:脱细胞猪皮基质材料在Ⅰ型胶原酶中20h降解率为(78±2.7)%,拉伸强度为789±14kPa;24h降解率为(92±2.3)%,样品已碎裂成小块儿;28h样品已基本降解完全.植入动物实验显示样品植入24周后可完全降解,表明猪皮细胞外基质具有良好的可降解性能,为其可控性降解研究提供相关依据.As a special natural biological derivative material,extracellular matrix (ECM) provides biophysical and mechanical support for cells and provides a good microenvironment for tissue regeneration. in vitrOand in vivOdegradation studies were performed herein on acellular porcine skin matrix material by treating with Type Ⅰ collagenase and by implanting the material intOanimals, respectively, and the degradation pattern and mechanical properties of the material were analyzed. Results showed that degradation rate of the acellular porcine skin matrix material treated in vitrOwith Type Ⅰ collagenase was (78±2.7)% at20 h,and the tensile strength was 789±14 kPa.The degradation rate reached (92±2.3)% at24 h ,and the sample was broken intOsmall pieces.At28 h, the sample was completely degraded.In addition, in vivOexperiments showed that the sample implanted intOanimals was completely degraded after24 weeks.All these results indicated that the extracellular matrix of porcine skin has good biodegradable properties, providing evidences for further study of its controllable degradation.
