摘 要:环境热应激是制约我国南方地区集约化家禽养殖业发展的一个重要因素,持续高温使蛋鸡出现产蛋率和蛋品质下降等问题,高温热应激是蛋鸡产业发展不能忽视的重要问题。该试验研究常温对照组和高温热应激组成年蛋鸡卵巢组织的差异mRNAs表达谱。60只26周龄成年蛋鸡随机分为常温对照组和高温热应激组,30d后2组分别随机选用3只共6只采集卵巢组织制备HE染色组织切片并利用深度测序技术分析mRNAs差异表达谱。相比对照组,高温组组织切片中卵泡的内容物增多,着色加深,卵泡壁增厚。测序数据统计分析发现,热应激处理后相比常温对照组有146个mRNAs表达水平差异显著(P<0.05),77个上调表达、69个下调表达;对表达差异基因功能分析发现,其中显著性差异(P<0.05)的调控通路有维生素B6代谢、卵母细胞减数分裂、柠檬酸代谢和细胞周期。以上研究结果为进一步解析蛋鸡卵巢热应激、提高蛋鸡生产性能奠定了基础。The heat stress caused by continuous high temperature weather has become an important restricting factor in the poultry industry. So, in this study, the differential expression profile of mRNAs in ovarian tissue of laying hens were investigated in normal temperature group and high temperature group. 60 adult laying hens with 26 weeks old were randomly divided into two groups, fed a same diet for 30 days respectively.Then the ovarian tissue of 6 hens from the three groups (3 from one group randomly) was collected to prepare HE stained tissue sections and be analyzed differential expression profiles of mRNAs by deep sequencing. In the section of ovarian tissue, compared with the control group, the contents of follicles were increased, the color was deepened, and the follicular wall was thickened in the high temperature group. The statistical analysis of sequencing data found that there were 146 mRNAs with the significant difference expression level ( P <0.05) in high temperature group compared with the normal control group;the function analysis of difference expressed mRNAs found that the regulation pathways related to the heat stress were vitamin B 6 metabolism, oocyte meiosis, citrate cycle and cell cycle.This research provided a foundation for further analyzing ovarian heat stress and improving performance of laying hens.
