摘  要:为提高樟子松种子园结实量,本研究采用染色法和培养基法测定不同贮藏条件下的花粉生活力,并通过扫描电镜进一步观察樟子松花粉形态特征。结果表明:I2-KI法测得花粉生活力最高,达到81.91%,I2-KI法最适于测定樟子松花粉生活力;在培养基中添加蔗糖浓度为10%和硼酸浓度为0.03%的培养基组合下,樟子松花粉萌发率最高,可达98.1%。扫描电镜观察樟子松花粉形态特征并测定樟子松花粉的极轴P,平均值为46.808μm;赤道长E,平均值为43.756μm,并计算P/E值,得出平均值为1.049 06。通过扫描电镜观察到樟子松花粉的表面粗糙,有很多的花粉囊,中间有一条极轴间区,把樟子松花粉分成两半。本研究结果为樟子松进行人工控制授粉提供理论基础,在林业生产上具有重要的指导意义。In order to improve seed yield of Pinus sylvestris var.mongholica Litv.seed orchard,the pollen viability under different storage conditions was measured by staining and culture media,and the pollen morphology characteristics were further observed by scanning electron microscopy in this study.The results showed that the I2-KI method had the highest pollen viability,which reached 81.91%.I2-KI method could be suitable for the determination of pollen viability.The germination rate of Pinus sylvestris var.mongholica Litv.pollen was the highest,reaching 98.1%,when sucrose concentration was 10%and boric acid concentration was 0.03%in the medium.Scanning electron microscopy was used to observe the morphological characteristics of the pollen of camphor pine and to measure the level axis P of the pollen,with an average value of 46.808μm.The mean value of equatorial length E was 43.756μm,and the P/E value was calculated.The average value was 1.049 06.The surface of Pinus sylvestris pollen was observed by scanning electron microscopy.There were many pollen sacs are very rough,and there was a region between the axles.The pollen was divided into two parts.The research results provide a theoretical basis for the artificial control pollination of Pinus sylvestris and has important guiding significance in forestry production.
