摘  要:目的分析3株salivirus病毒基因组特征,为了解我国salivirus病毒分子进化特征提供数据。方法对2018年扬州市儿童腹泻病例中3份salivirus病毒检测阳性的样本核酸进行高通量测序,获得该病毒的全基因组,应用MEGA 7.0软件构建系统进化树,通过MegAlign软件对核苷酸及编码区氨基酸序列进行同源性分析。结果 3株salivirus病毒均属于salivirus A1亚型,病毒之间基因组核苷酸及编码区氨基酸同源性均为99.4%~99.7%。与匈牙利毒株Hu/Baja-5332/2013/HUN/Salivirus A的同源性最高,核苷酸和编码区氨基酸同源性分别为96.5%~96.7%及98.0%~98.1%,处于同一进化分支。3株salivirus与国内salivirus A1亚型毒株的核苷酸和编码区氨基酸同源性分别为94.2%~95.2%及95.2%~97.1%;与国外salivirus A1亚型的核苷酸和编码区氨基酸的同源性分别为89.7%~96.7%及95.0%~98.5%,与国外salivirus A2亚型的核苷酸和编码区氨基酸的同源性分别为81.8%~81.9%及88.3%~88.4%。结论 3株salivirus病毒遗传进化特征不同于我国已报道的salivirus病毒,丰富了我国salivirus病毒的基因数据库。Objective To study the genomic characteristics of three strains of salivirus in Yangzhou city of China. Methods The genome of the three strains of salivirus isolated from diarrhea cases in 2018were obtained by high-throughput sequencing.Phylogenetic tree was constructed by MEGA 7.0software.Homology analysis of nucleotide and amino acids were conducted by MegAlign software. Results The three strains of salivirus belong to the salivirus A1subtype;the homology of nucleotides and the coding region amino acids were both 99.4%-99.7%.The sequences showed highest homology with that of Hungary strain Hu/baja- 5332/2013/HUN/Salivirus A.The homology of nucleotides and coding region amino acids were 96.5%-96.7% and 98.0%-98.1%,respectively.Phylogenitic anylsis showed they were in the same evolutionary branch.The nucleotide and coding amino acid homology of three strains of salivirus compared with China reference strains of salivirus A1subtype were 94.2%-95.2% and 95.2%-97.1%,respectively;89.7%-96.7% and 95.0%- 98.5%,respectively when compared with foreign salivirus A1subtype;81.8%-81.9% and 88.3%-88.4%,respectively when compared with salivirus A2. Conclusions The genetic and evolutionary characteristics of three strains of salivirus isolated in Yangzhou city of China are different from those reported previously in China.
