四川省首例输入性D8基因型麻疹病毒基因特征分析

摘　　要：目的对四川省1例输入性麻疹病例的病毒株进行基因特征分析。方法采用Vero/SLAM细胞分离培养麻疹病毒,通过逆转录-聚合酶链反应(RT-PCR)方法扩增麻疹病毒N基因羧基末端634个核苷酸片段,并对PCR产物进行序列测定和同源性分析。结果该麻疹病毒分离株与世界卫生组织(WHO)D8基因型参考株在基因亲缘性关系树上同属1个分支,核苷酸和氨基酸同源性分别为98.4%和98.0%。结论该病例为四川省首例输入性D8基因型麻疹病例。Objective To analyze the genetic characteristics of an imported measles virus strain isolated in Sichuan province of China. Methods Throat swab sample was isolated and cultured by Vero/SLAM cells.A 634bp fragment of the N gene was amplified by reverse transcription-polymerase chain reaction(RT-PCR) and subjected to sequence and homological analysis. Results The isolate was clustered within the same genotype group of the WHO genotype D8reference strain.The nucleotide and amino acid homology between them were 98.4%and 98.0%,respectively. Conclusions The newly imported measles virus strain in Sichuan of China is genotype D8.
