摘 要:目的探讨真空离心浓缩DNA样本技术在法医学中的应用。方法 2800M(1ng/μl)、007(0.05 ng/μl)、9947A(0.005、0.01、0.02、0.04 ng/μl)同时在45℃的真空离心浓缩仪中浓缩5倍,观察图谱变化以及样本间是否有污染;3组各10份007(0.05ng/μl)分别在30℃、45℃、60℃浓缩5倍,比较不同温度浓缩获得的图谱间差异;现场物证DNA模板48份,在45℃浓缩4.4倍至7.7倍。对上述浓缩后模板使用GlobalFilerTM试剂盒扩增、检测。结果 2800M、007、9947A均获得正确STR分型,未发现相互污染,低浓度样品图谱质量明显改善;007在不同温度浓缩,获得图谱的平均峰高有差异;现场物证模板浓缩后图谱质量有显著改善,检出率提高。结论真空离心浓缩技术可以应用于法医DNA样本浓缩,提高微量疑难检材的检验成功率。Objective To investigate the application of vacuum centrifugal concentrated DNA sample technology in forensic science. Methods 2800 M(1 ng/μl)、007(0.05 ng/μl)、9947 A(0.005、0.01、0.02、0.04 ng/μl) were concentrated to 5 times by the vacuum centrifugal concentrator in 45 ℃, the STR change and the contamination between the samples were observed;3 groups 007(0.05 ng/μl) each of which has 10 samples were concentrated to 5 times in 30 ℃, 45 ℃, 60 ℃ respectively and the differences by different temperature concentration were compared;48 DNA samples of the crime scene concentrated 4.4 times to 7.7 times in 45 ℃ were amplified using GlobalFilerTM kit and tested. Results Both 2800 M, 007, 9947 A obtained the correct STR typing, did not find mutual pollution, and significantly improved the quality of low concentration samples;The average peak height of STR varies in 007 concentration at different temperatures, The quality of STR typing was improved significantly after the concentration of the crime scene samples, and the success rate was improved. Conclusion Vacuum centrifugal concentration technology can be applied to the concentration of forensic DNA samples to improve the success rate of trace samples.
