摘 要:目的研究黑河流域中下游黑果枸杞16个野生居群的遗传多样性,为黑果枸杞的遗传育种及保护机制研究提供依据。方法在建立ISSR分子标记的基础上,人工读带确定引物位点,利用Popgen32软件分析黑果枸杞的多态性、遗传相似系数及遗传距离。使用NTSYS软件建立黑果枸杞的遗传距离聚类图。结果采用6个ISSR引物对16个黑果枸杞居群的基因组DNA进行扩增后共检测到126个位点,多态位点百分率为92.86%,不同居群间的多态位点百分率范围为6.35%~50.00%。Nei’s基因多样性指数H为0.2228,Shannon信息指数I为0.3459,分析得出居群间的变异为41.85%。聚类的遗传距离介于0.03~0.16,大体聚为2类。结论黑河流域中下游黑果枸杞在种群水平上遗传多样性丰富,对环境的适应能力较强;黑果枸杞有复杂的遗传背景,且与地理来源相关性不强。Objective To study the genetic diversity of 16 wild populations of Lycium ruthenicum Murr.;To provide a theoretical basis for genetic breeding and establishment of protective mechanisms of Lycium ruthenicum Murr..Methods Based on the establishment of ISSR molecular markers,the artificial reading bands were used to identify the primer sites.The polymorphisms,genetic similarity coefficients and genetic distances of Lycium ruthenicum Murr.were analyzed using Popgen32 software.NTSYS software was used to establish a genetic distance clustering map of Lycium ruthenicum Murr..Results Totally 6 ISSR primers were used to amplify the genomic DNA of 16 populations and a total of 126 sites were detected.The percentage of polymorphic sites was 92.86%.The percentage of polymorphic sites among different populations was in the range of 6.35%–50.00%.The Nei’s gene diversity index H was 0.222 8 and Shannon’s information index I was 0.345 9.The analysis showed that the variation among populations was 41.85%.The genetic distance of clustering was in the range of 0.03–0.16,which could roughly be divided into two categories.Conclusion The genetic diversity of Lycium ruthenicum Murr.in middle and lower reaches of Heihe River basin is abundant at the population level,and has a strong adaptability to the environment.Lycium ruthenicum Murr.has a complex genetic background and is not strongly related to geographical origin.
