摘 要:目的:建立快速检测结核杆菌菌液浓度的方法。方法:制备土豆鸡蛋培养基,以卡介苗为菌种,培养结核杆菌并制成一定浓度的菌液,紫外可见光分光光度计200-500nm波长扫描,选择检测最佳波长;用最佳波长检测系列浓度结核杆菌菌液浓度,考察菌液浓度与菌液吸光度之间的关系,建立标准曲线。结果:在295nm波长下,结核杆菌菌液浓度与菌液吸光度之间具有良好的相关性,相关系数r=1(P<0.01);直线回归方程为Y=-4.9×10^5+1.7×10^6X。结论:紫外分光光度法可用于快速准确检测结核杆菌菌液浓度。Objective: To establish a rapid method for the detection of Mycobacterium tuberculosis solution.Methods:Potato- egg culture medium was prepared,Mycobacterium tuberculosis was cultured and various concentration of bacterial solution was made.200-500nm wavelength scanning with UV-vis spectrophotometer was used to select the best wavelength for detection.The optimum wavelength was used to detect the concentration of Mycobacterium tuberculosis solution,the relationship between the concentration of bacterial solution and the absorbance of bacterial solution was investigated,and the standard curve was established.Results:At 295nm wavelength,there was a good correlation between the concentration of Mycobacterium tuberculosis solution and the absorbance of Mycobacterium tuberculosis solution,the correlation coefficient r=1( P <0.01),and the linear regression equation was Y=-4.9×10^ 5+1.7×10^ 6X.Conclusion:UV spectrophotometry can be used for rapid and accurate detection of Mycobacterium tuberculosis solution.
