摘 要:Klotho是新发现的抗衰老因子,主要表达于肾脏,对肾脏而言具有广泛的保护作用,其中对肾间质纤维化(RIF)的抑制作用是Klotho发挥肾脏保护作用的一个重要机制。Klotho可通过调控转化生长因子-β1(TGF-β1)、Wnt/β-联蛋白(β-catenin)、肾小管上皮细胞间质转化(EMT)、肾素-血管紧张素系统(RAS)、成纤维细胞生长因子2(FGF-2)、内质网应激(ERS)等多种与RIF相关的信号通路,多靶点抑制RIF的发生发展。因此,Klotho被认为是新的RIF抑制因子,以Klotho为干预靶点,可以为将来RIF的防治提供新思路。Klotho is recently identified as an anti-aging factor and it is mainly located in the kidney and has exhibited various renal-protective effects including inhibition of renal interstitial fibrosis( RIF). Nowadays Klotho is believed to inhibit RIF by targeting transforming growth factor β1( TGF-β1),Wnt/β-catenin,epithelial-mesenchymal transition( EMT),reninangiotensin system( RAS),fibroblast growth factor-2( FGF-2) and endoplasmic reticulum stress( ERS) signaling involved in RIF. Klotho has been regarded as a novel factor for RIF inhibition and targeting Klotho may provide new ideas for the prevention and treatment of RIF in future.
