摘  要:目的:建立三痹汤中主要成分龙胆苦苷含量测定的方法,为三痹汤的质量控制提供实验依据。方法:采用HPLC-DAD法测定三痹汤中龙胆苦苷的含量,仪器为安捷伦1260infinityⅡ,色谱柱为Inetsil-ODSC18(4.6mm×250mm,5μm),流动相为乙腈(A)-0.05%磷酸水溶液(B)进行梯度洗脱,流速为1mL/min,检测波长为250nm。结果:该方法的精密度,稳定性符合要求,主要成分的峰面积和质量浓度之间的线性关系良好,r大于0.999,平均加样回收率99.8%~100.0%,RSD小于0.1%,龙胆苦苷的仪器检测限为0.005483μg/mL,定量限为0.01845μg/mL。结论:本文建立了三痹汤的HPLC主要成分龙胆苦苷含量测定方法,可以为三痹汤质量控制提供实验依据参考。Objective: To developa method for determination of gentiopicroside in San Bi Tangextracts,provide experimental basis for quality control of San Bi Tanggranules.Methods:In this study, the gradient elution of acetonitrile (A)- water (B) was carried out by using the method of HPLC-DAD with Agilent 1260 infinity II as the instrument, Inetsil- ODS C18 (4.6 mm×250 mm, 5 μm) as the column, and the mobile phase was acetonitrile (A)- water (B). The flow rate was 1 mL/min, and the detection wavelength was 250 nm.Results:The precision and stability of the method meet the requirements. The linear relationship between peak area and mass concentration of main components is good, R is greater than 0.999. The average recovery of sample addition is 99.8%~ 100.0%, RSD is less than 0.1%, the detection limit of gentiopicroside is 0.005483μg/mL, and the quantitative limit is 0.01845μg/mL.Conclusions: A method for the determination of gentiopicrosidein the extract of San Bi Tang has been established, which can provide experimental basis for the quality control of San Bi Tang.
