铃兰高效繁育体系条件探究

摘  要:为了解决铃兰组培过程中的污染问题,构建高效繁育体系,采用铃兰不同器官作为外植体,以愈伤组织形成率及污染率作为评价指标,比较培养基配方、抑菌剂种类和浓度对愈伤组织诱导及组培污染菌的影响,筛选出铃兰高效繁育体系。结果表明:(1)不同外植体诱导愈伤组织的能力不同,对激素的要求也有所不同,铃兰嫩芽配合附加0.3mg/L6-BA和0.2mg/LNAA的MS培养基组合对愈伤组织的诱导效果最好;(2)浓度为0.20g/L的代森锰锌能够有效的抑制组培污染菌的形成,污染率可降低至4.70%,同时对愈伤组织的形成具有促进作用,是最佳的抑菌剂选择。In order to solve the pollution problems in the process of lilies of the valley transformation, construct efficient breeding system, adopts the lilies of the different organs as explant, callus formation rate and the pollution rate as evaluation index, and comparative culture medium formula, bacteriostatic agent type and concentration of callus induction and the influence of contaminated bacteria, lilies of the efficient breeding system. The results showed that :(1) Different explants had different ability to induce callus and different requirements for hormones. The best effect of callus induction was achieved by combining the buds of lilies of the lilies with MS medium supplemented with 0.3 mg/L 6-BA and 0.2 mg/L NAA;(2) Mancozeb with a concentration of 0.20g/L can effectively inhibit the formation of tissue culture contaminated bacteria, and the pollution rate can be reduced to 4.70%. At the same time, it can promote the formation of callus and is the best choice of bacteriostatic agent.
