摘 要:目的探讨下丘脑弓状核促性腺激素释放激素(Gn RH)、Kisspeptin和Kiss1r蛋白和m RNA在多囊卵巢综合征(PCOS)大鼠模型中的表达水平。方法取24只雌性Wistar大鼠,随机分为对照组(正常大鼠)和实验组(应用来曲唑建立PCOS大鼠模型),两组各12只。对照组给予等量羧甲基纤维素溶液灌胃。实验组应用来曲唑1 mg/kg联合1%羧甲基纤维素溶液进行每日灌胃,连续3周。经苏木精-伊红染色观察两组大鼠卵巢组织病理学改变,检测内分泌激素水平,分别采用免疫印迹法与实时荧光定量PCR法对大鼠下丘脑弓状核Gn RH、Kisspeptin、Kiss1r蛋白和m RNA表达量进行检测。结果实验组血清卵泡刺激素(FSH)、睾酮(T)和黄体生成素(LH)的水平较对照组显著升高(P<0.01)。实验组Kisspeptin、Kiss1r蛋白和m RNA表达量较对照组显著降低(P<0.05),Gn RH蛋白和m RNA表达量较对照组显著升高(P<0.01)。结论PCOS大鼠Gn RH表达异常升高,Kisspeptin表达异常降低,提示大鼠下丘脑弓状核Kisspeptin/Kiss1r系统表达异常,与PCOS的神经内分泌密切相关,可能在PCOS的病理过程中起到重要作用。Objective To investigate the expression levels of gonadotropin releasing hormone(GnRH),Kisspeptin and Kiss1r protein and mRNA in the polycystic ovary syndrome(PCOS)rat model.Methods 24 female Wistar rats were randomly divided into control group(normal rats)and experimental group(letrozole was used to establish PCOS rats model),12 rats in each group.The control group was given an equal amount of carboxymethylcellulose solution for intragastric administration.The experimental group was treated with letrozole 1 mg/kg in combination with 1%carboxymethylcellulose solution for 3 weeks.The histopathological changes of the ovary of the two groups were observed by hematoxylin-eosin staining,and the levels of endocrine hormones were detected.The expressions of GnRH,Kisspeptin,Kiss1r protein and mRNA in the hypothalamus arch nucleus of the rats were detected by western blot and real-time fluorescence quantitative PCR,respectively.Results The levels of follicle stimulating hormone(FSH),testosterone(T)and luteinizing hormone(LH)in the experimental group were significantly higher than those in the control group(P<0.01).The expression levels of Kisspeptin,Kiss1r protein and mRNA in the experimental group were significantly lower than those in the control group(P<0.05),and the expression levels of GnRH protein and mRNA were significantly higher than those in the control group(P<0.01).Conclusion The abnormal increase of GnRH expression and the abnormal decrease of Kisspeptin expression in PCOS rats suggest that the abnormal expression of Kisspeptin/Kiss1r system in the hypothalamic arcuate nucleus of rats is closely related to the neuroendocrine of PCOS and may play an important role in the pathological process of PCOS.
