摘  要:目的探讨热休克蛋白27(Hsp27)在超低温冷冻大鼠精子中的表达特征,分析其与精子活力的相关性。方法选择20只性成熟雄性大鼠20只,麻醉状态下获得精子悬液,检测精子活力。根据精子活力将大鼠分为精子活力≥50%组(12只)和<50%组(8只),之后进行超低温冷冻试验。分别检测冷冻前后Hsp27表达水平,分析精子活力与Hsp27表达水平相关性以及影响冷冻后大鼠精子活力的因素。结果大鼠精子经超低温冷冻后精子总活力和Hsp27蛋白表达均出现明显下降[(68.12±4.62)%vs.(42.52±3.73)%、(11.62±3.51)vs.(3.85±1.35),P<0.05];精子活力≥50%组大鼠精子经冷冻前后精子活力大于精子活力<50%组[(67.52±4.75)vs.(40.54±3.53)、(38.62±4.51)vs.(21.85±1.05),P<0.05]。精子活力≥50%组、精子活力<50%组精子活力与Hsp27蛋白表达均呈正相关(r=0.421,0.458,P<0.05),Logistic回归分析示Hsp27蛋白表达水平与超低温冷冻后精子活力独立相关(β=0.721,OR=2.013)。结论Hsp27在超低温冷冻大鼠精子中表达下调,其表达水平与精子活力相关,Hsp27表达下降可能是冷冻后精力活力下降的可能机制。Objective To investigate the expression of heat shock protein 27(Hsp27)in sperm of cryopreserved rats,and to analyze its correlation with sperm motility.Methods 20 sexually mature male rats were selected.Sperm suspension was obtained under anesthesia and sperm motility was measured.According to sperm motility,the rats were divided into groups with sperm motility(>50%)and<50%(8),and then cryopreservation test was carried out.The expression levels of Hsp27 were detected before and after cryopreservation,and the correlation between sperm viability and expression level of Hsp27 was analyzed,as well as the factors affecting sperm viability after image cryopreservation.Results The sperm motility and Hsp27 protein expression of rat sperm decreased significantly after ultra-low temperature freezing[(68.12±4.62)%vs.(42.52±3.73)%,(11.62±3.51)vs.(3.85±1.35),P<0.05],Sperm vitality≥50%group sperm sperm before and after freezing than the sperm activity<50%group[(67.52±4.75)vs.(40.54±3.53),(38.62±4.51)vs.(21.85±1.05),P<0.05].There was a positive correlation between sperm motility≥50%group and sperm motility<50%group and Hsp27 protein expression(r=0.421,0.458,P<0.05).Logistic regression analysis showed that Hsp27 protein expression level was independently associated with sperm motility after cryopreservation(β=0.721,OR=2.013).Conclusion The expression of Hsp27 in cryopreserved rat sperm is down-regulated.The expression level of Hsp27 is closely related to sperm viability.The decrease of Hsp27 expression may be the possible mechanism of the decrease of sperm viability after cryopreservation.
