摘  要:目的探讨川芎嗪对糖尿病肾病(DN)大鼠肾组织糖原合成酶激酶(GSK-3β)、血管内皮生长因子(VEGF)及细胞间黏附分子-1(ICAM-1)表达的影响。方法将30只SD大鼠随机分为对照组、模型组和川芎嗪组各10只,模型组和川芎嗪组饲喂高脂高糖饮食,腹腔注射小剂量链脲佐菌素(25mg/kg)建立DN大鼠模型。川芎嗪组灌胃150mg/(kg·d)的川芎嗪,1次/d,持续8周,模型组和对照组分别灌胃等量的生理盐水。于治疗前后,测定各组大鼠空腹血糖(FBG);治疗后,测定肾功能相关指标血肌酐(SCr)、尿素氮(BUN)的水平和24h尿蛋白含量;采用WesternBlot测定GSK-3β、VEGF和ICAM-1的蛋白表达水平;HE染色观察肾脏组织病理变化。结果与对照组相比,模型组和川芎嗪组大鼠FBG显著升高(P<0.05),肾功能相关指标SCr、BUN和24h尿蛋白含量显著升高(P<0.05),GSK-3β、VEGF和ICAM-1的蛋白表达显著升高(P<0.05);与模型组相比,川芎嗪组大鼠FBG显著降低(P<0.05),肾功能相关指标SCr、BUN和24h尿蛋白含量显著降低(P<0.05),GSK-3β、VEGF和ICAM-1的蛋白表达显著下降(P<0.05),HE染色显示肾脏病理变化明显减轻。结论川芎嗪可能通过抑制GSK-3β、VEGF和ICAM-1的表达,改善DN大鼠的肾脏功能。Objective To explore the effects of ligustrazine on the expression of glycogen synthase kinase(GSK-3β),vascular endothelial growth factor(VEGF)and intercellular adhesion molecule-1(ICAM-1)in renal tissues of rats with diabetic nephropathy(DN).Methods Thirty SD rats were randomly divided into control group,model group and ligustrazine group,with 10 rats in each group.Model group and ligustrazine group were fed a high-fat and high-glucose diet,and intraperitoneally injected low-dose streptozotocin(25 mg/kg)to establish DN rat models.Ligustrazine group was intragastrically administered with 150 mg/(kg·d)of ligustrazine once/d for 8 w,and model group and control group were intragastrically administered with the same amount of normal saline.The fasting blood glucose(FBG)was measured in each group before and after treatment.After treatment,related indexes of renal function of serum creatinine(SCr),urea nitrogen(BUN)and 24 h urine protein were measured.Western Blot was used to measure the protein expression levels of GSK-3β,VEGF and ICAM-1.Renal tissue pathological changes were observed by HE staining.Results Compared with control group,the FBG in model group and ligustrazine group was significantly increased(P<0.05),and the renal function related indexes SCr,BUN and 24 h urine protein were significantly increased(P<0.05),and the protein expression levels of GSK-3β,VEGF and ICAM-1 were significantly increased(P<0.05).Compared with model group,the FBG in ligustrazine group was significantly decreased(P<0.05),and the renal function related indexes SCr,BUN and 24 h urine protein were significantly decreased,and the protein expression levels of GSK-3β,VEGF and ICAM-1 were significantly decreased(P<0.05),and HE staining showed a significant reduction in renal pathological changes.Conclusion Ligustrazine may improve renal function in DN rats by inhibiting the expression of GSK-3β,VEGF and ICAM-1.
