摘 要:目的 观察扶正固虚、清热解毒联合疗法对小鼠肝癌的治疗作用,并探讨其机制.方法 48只小鼠均制备Hca-F肝癌模型,后随机分为4组各12只,于第2、4、6、8、10、12天腹腔注射药物或生理盐水.联合治疗组给予15 mL/kg扶正固虚药物+15 mL/kg清热解毒药物,扶正固虚组腹腔给予30 mL/kg扶正固虚药物,清热解毒组给予30 mL/kg清热解毒药物,肿瘤对照组给予0.5 mL生理盐水;另选取12只健康小鼠(设定为空白对照组),给予0.5 mL生理盐水.处死后称量各组小鼠肿瘤重量,并计算抑瘤率(除外空白对照组);流式细胞技术检测各组外周血T淋巴细胞亚群CD4+T细胞、CD8+T细胞百分比,计算CD4+T/CD8+T;ELISA法检测各组血清IL-2、IL-6、IFN-γ、TNF-α.结果 与肿瘤对照组比较,联合治疗组、扶正固虚组、清热解毒组肿瘤重量明显降低,CD4+T细胞、CD8+T细胞百分比及IL-2、IL-6、IFN-γ、TNF-α水平升高,CD4+T/CD8+T细胞显著下降,尤以联合治疗组效果最为明显(P均<0.05).结论 扶正固虚、清热解毒联合疗法对小鼠肝癌有治疗作用,机制可能与其可调节T淋巴细胞亚群平衡,促进细胞因子分泌,从而提升免疫力有关.Objective To investigate the effect and mechanism of strengthening the body resistance ( SBR) therapy combined with heat-clearing and detoxifying ( HCD) therapy in the treatment of mice with liver cancer. Methods Fortyeight mouse models with Hca-F liver cancer were constructed successfully and randomly divided into the tumor control group, SBR group, HCD group and combination group, with 12 in each. They were administered with drugs or normal saline on day 2,4,6,8,10, and 12. Mice in the combination group received 15 mL/kg SBR medicine plus 15 mL/kg HCD medicine. Mice in the SBR group received 30 mL/kg SBR medicine. Mice in the HCD group received 30 mL/kg HCD medicine. Mice in the tumor control group received 0. 5 mL normal saline. Twelve healthy mice were selected as the blank control with administration of 0. 5 mL normal saline. Tumor weights were measured after execution. Tumor inhibitory rate was calculated. The percentages of CD4 + T cells, CD8 + T cells in the peripheral blood T lymphocytes and the CD4 + T/ CD8 +T ratio were detected by flow cytometry. In addition, the content of interleukin 2 (IL-2), IL-6, interferon-γ(IFN-γ), and tumor necrosis factor-α(TNF-α) were measured by ELISA. Results Compared with the tumor control group, the tumor weights decreased in the other treatment groups. The percentages of CD4 T cells and CD8 T cells,levels of IL- 2, IL-6, IFN-γ, and TNF-α increased, while CD4 + T/CD8+ T decreased significantly in the other treatment groups, with the best effect in the combination group ( all P <0.05). Conclusion Combination of SBR and HCD has anti-tumor effect by regulating the balance of T lymphocyte subsets in liver cancer mice, increasing the levels of cytokines, and thus enhancing immunity.
