摘 要:目的通过对比正常皮肤与气虚皮肤的药物经皮渗透情况,以探讨气虚证对药物透皮吸收的影响。方法采用游泳疲劳法制造气虚模型,观察并对比正常大鼠与气虚大鼠的外观表现;通过皮肤组织观察,对比正常皮肤与气虚皮肤的特点;以延胡索乙素为模型药物进行透皮实验,测量2、4、8、12、24h时接收液的药物浓度,计算累积渗透量和渗透系数。结果气虚大鼠在外观方面与正常大鼠有明显区别;延胡索乙素在气虚大鼠离体皮肤上,不同时间的累积渗透量与正常组大鼠相比均减少,在气虚大鼠皮肤及正常大鼠皮肤上的渗透系数分别为(97.6±8.6)μg/(cm^2·h)和(188.3±10.7)μg/(cm^2·h),气虚组的渗透系数显著低于正常组(P<0.01)。结论气虚大鼠的皮肤对延胡索乙素的渗透有显著的阻滞作用。Objective To study the effect of Qi deficiency syndrome on drug transdermal absorption by comparing the transdermal permeation of drugs between normal skin and Qi deficiency skin.Methods The Qi-deficiency model was made by swimming fatigue method,and the appearance of normal rats and Qi-deficiency rats was observed and compared.The characteristics of normal skin and Qi-deficiency skin was compared by skin tissue observation.The transdermal experiment was carried out with tetrahydropalmatine as model drug,and the drug concentration of the solution was determined at 2,4,8,12 and 24 hours,and the cumulative permeability and permeability coefficient were calculated.Results There was a significant difference in appearance between Qi-deficiency rats and normal rats.The accumulative permeation amount of tetrahydropalmatine on the isolated skin of rats with Qi deficiency at different time decreased compared with that of normal rats.The permeation coefficient of tetrahydropalmatine on the skin of rats with Qi deficiency and normal rats was(97.6±8.6)μg/(cm^2·h)and(188.3±10.7)μg/(cm^2·h)respectively.The permeability coefficient of Qi deficiency group was significantly lower than that of normal group(P<0.01).Conclusion The skin of rats with Qi deficiency has a significant blocking effect on the penetration of tetrahydropalmatine.
