摘  要:目的:探讨不同浓度的二氢青蒿素(DHA)对肝癌HepG-2细胞生长的影响,以及影响其增殖可能的作用机制。方法:体外培养人肝癌细胞HepG-2,将实验分成加药实验组和空白对照组。加药组加入不同浓度的二氢青蒿素(25μM、50μM、100μM、150μM、200μM)干预。利用CCK8法检测不同浓度的二氢青蒿素对肝癌HepG-2细胞增殖抑制的情况,采用Westernblot检测细胞内PTEN/Akt信号通路中PTEN、Akt、CyclinD1、P27相关蛋白的表达情况。结果:由CCK8的结果得出的结果,二氢青蒿素可以抑制肝癌HepG-2细胞的生长,并呈一定的浓度和时间依赖性抑制。二氢青蒿素处理48h后,通过Westernblot的结果显示,PTEN、P27表达量升高,CyclinD1和Akt表达量降低(P<0.05)。结论:二氢青蒿素抑制肝癌细胞HepG-2的增殖可能与抑制PTEN/Akt信号通路,阻滞细胞生长周期有关。OBJECTIVE: To investigate the effects of different concentrations of dihydroartemisinin (DHA) on the growth of HepG-2 cells and the possible mechanisms of their proliferation. METHODS: HepG-2 cells were cultured in vitro, and the experiment was divided into a dosing experiment group and a blank control group. The drug-treated group was treated with different concentrations of dihydroartemisinin (25 μM, 50 μM, 100 μM, 150 μM, 200 μM). CCK8 method was used to detect the inhibition of proliferation of HepG-2 cells by different concentrations of dihydroartemisinin. The expressions of PTEN, Akt, CyclinD1 and P27 related proteins in PTEN/Akt signaling pathway were detected by Western blot. RESULTS: Dihydroartemisinin inhibited the growth of HepG-2 cells in a concentration- and time-dependent manner as a result of CCK8. After treatment with dihydroartemisinin for 48h, the expression of PTEN and P27 was increased, and the expression of CyclinD1 and Akt was decreased (P<0.05). Conclusion: Dihydroartemisinin inhibits the proliferation of HepG-2 cells in hepatocellular carcinoma cells, which may be related to the inhibition of PTEN/Akt signaling pathway and the arrest of cell growth cycle.
