摘  要:本研究旨在探讨人参浸出液(Rs)对铅暴露鲤鱼卵巢细胞(GCO)减毒作用。以GCO 细胞为研究对象,采用四甲基偶氮唑盐(MTT)法,以0.01 mmol/L Pb 预处理细胞1 h 后,添加不同浓度人参浸出液(2%Rs、4%Rs、6%Rs、8%Rs)进行治疗后,在12、24、48 h 检测细胞活性;48 h 检测细胞超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)和过氧化氢酶(CAT)指标和金属硫蛋白基因(MT)相对表达量。结果表明,不同浓度Rs 添加能够提高 GCO 细胞活性;不同浓度Rs 添加能够缓解 Pb 诱导 GCO 细胞毒性,显著提高抗氧化能力(SOD、CAT、GSH-Px),调节 MT 基因表达,4%Rs 添加时最佳。综上所述,不同浓度Rs 能够缓解 Pb 诱导 GCO 细胞毒性,4%Rs 添加时最佳。This study was to investigate the attenuating effect of ginseng extract(Rs) on lead-exposed grass carp ovary cells(GCO). In this study, using GCO as object and Pb as inducer of toxic, the effects of different dose of Rs(2%,4%, 6% and 8%) on cell viability followed 0.01 mmol/L challenge were measured by methyl thiazolyl tetrazolium(MTT) method at 12 h, 24 h and 48 h. The relative expression of metallothionein(MT) gene, SOD, GPx and CAT were measured at 48 h. The results showed that different concentrations of Rs could increase the activity of GCO cells. The different concentrations of Rs could alleviate the Pb-induced GCO cytotoxicity, including significantly increased the antioxidant capacity(SOD, CAT, GPx) and regulated MT gene expression. The best addition is 4%Rs. Thus, the different concentrations of Rs can alleviate Pb-induced GCO cytotoxicity and the best addition is 4% Rs.
