摘　　要：目的 :研究TGF - β对成骨细胞增殖和分化的影响。方法 :利用已建立的胎兔颅骨成骨样细胞体外模型 ,采用MTT法、流式细胞仪检测、骨钙素、胶原和钙含量测定来分析不同浓度的TGF - β对成骨样细胞增殖和分化的影响。结果 :低浓度TGF - β(0 .0 1 0 .1ng/ml)促进成骨样细胞增殖 ,使S期细胞增加 ,但高浓度TGF - β(1 1 0ng/ml)抑制成骨样细胞的增殖。在TGF - β 0 .0 1～ 1ng/ml范围内 ,与对照组相比 ,TGF - β各治疗组的骨钙素、胶原和钙含量明显增加 ,并随TGF- β浓度增加而增加 ,到TGF - β 1 0ng/ml时 ,增加不再明显。结论 :TGF - β对成骨细胞的增殖表现为双相作用 ,低浓度促进 ,高浓度时抑制 ,可明显促进成骨细胞的分化 ,并呈一定的量效关系 ,其有效剂量为 0 .0 1 1ng/ml。Objective: To study the effects of transforming growth factor beta(TGF-β)on the proliferation and differentiation of primary cultured osteoblast like cells derived from fetal rabbit calvarium in vitro.Methods:The culture models of fetal rabbit calvarium osteoblast like cells in vitro was established and treated wth different concentrations of TGF-β.The MTT assay,flow cytomety,and osteocalcin(OCN),as well as collagen and calcium contents were used to observe the effects of TGF-β on proliferation and differentiation of osteoblast like cells.Results:The lower concentration of TGF-β(0.01～0.1ng/ml)enhanced osteoblast like cells proliferating,and caused the osteoblast like cells of S phase to increase,but the higher concentration of TGF-β (1～10ng/ml)inhibited osteoblast like cells to proliferate.Compared with control group,there were significant increases of OCN,collagen and calcium contents of osteoblast like cells in the groups receiving TGF-β and this increase accompanied with increased dose of TGF-β when dose in the rang of 0.01ng/ml to 1ng/ml were used,whereas no significant increases were found when a doses of 10ng/ml was applied.Conclusions:The effects of TGF-β on the proliferation of osteoblast like cells is biphasic and depends on the TGF-β concentration,including inhibition at high concentration and enhancement at the low concentration.TGF-β has significant promoting effect on differentiation of osteoblast like cells in a dose dependent manner,and its effective concentration is 0.01～1ng/ml.
