摘  要:目的:从大白兔胎肾细胞内提取、部分纯化治疗急性肾功能衰竭(ARF)的有效物质。方法:制备胎肾内生长刺激物质,经过盐析、透析、液相色谱分离提取、部分纯化,进行动物实验,3H—Tdr参入测定其CPM值。结果:胎肾细胞移植治疗ARF的有效成分是分子量为28 000的耐热蛋白质。70%以上的硫酸胺能沉淀其活性成分,离子交换色谱Ⅳ、Ⅴ峰为活性峰,凝胶过滤色谱Ⅰ、Ⅱ峰为活性峰。结论:胎肾细胞移植治疗ARF的有效成分是一种分子量为28 000,耐热的具有活性的细胞因子,这种因子可能仅存在胎肾内或只有胎肾内含量最高。Purpose: To extract and purify effective substance in foetus renal cells from large white rabbits, which could cure acute renal failure (ARF). Method;Growth stimulant in foetus renal cells was extracted and purified by salting out method, dialysis and liquid chromatography (LC). Activity was determined by animal experiment. CPM value was determined by 3H-TdR incorporation. Result: The effective composition which could cure ARF in foetus renal cells was an thermostabile protein of molecular weight 28KD. The active mass were precipitated in 70% ammonium sulfate. The active peaks were peak JY > V in ion-exchange chromatography and peak I , D in gel filtration chromatography. Conclusion: The effective composition which could cure ARF in foetus renal cells was an active thermostabile cellular factor which might exist only in foetus renal cells or had maximum content only in foetus renal cells.
