摘　　要：目的 :探讨汉坦病毒 (HTNV)感染诱导乳鼠脑组织热休克蛋白 (HSPs)表达及热休克蛋白与病毒蛋白的相互关系。方法 :采用免疫荧光法检测实验性乳鼠感染脑组织中病毒抗原及HSP70的表达 ,用ELISA、免疫印迹、免疫共沉淀方法分析病毒抗原和HSP70的表达。结果 :HTNV感染诱导了乳鼠脑组织神经细胞高表达HSP70 ,组织中HTNV NP与HSP70关系密切 ,免疫共沉淀结果显示HSP70与HTNV NP结合形成复合物。结论 :HTNV实验性乳鼠感染脑组织中高表达的HSP70与病毒核心抗原 (HTNV NP)形成非共价复合物 ,该复合物可能在病毒感染复制及宿主免疫反应中发挥作用。Purpose To investigate the interaction between heat shock proteins and viral components during viral assembly in the brain of suckling mice infected experimentally with Hantanvirus. Methods The Hantanvirus infection, HSP70 induction were demonstrated by immunofluorescence. The possible interaction between HTNV structural proteins and HSP70 was further analyzed by immunoprecipitation and demonstrated by SDS PAGE and Western Blot. Results HTNV infection caused a increased HSP70 expression at the cytoplasms of neurons in the brains of suckling mice. Monoclonal antibody (McAb) against HSP70 could coimmunoprecipitate the HTNV NP, and similarly McAb against HTNV NP could coimmunoprecipitate HSP70 from virus infected tissue extracts, but not from mock infected tissue extracts. Antibody, which recognizes one of these two proteins, does not cross react with the other. Thus, the association between HSP70 and HTNV NP appears specific. Conclusions Hantanvirus infection can infect experimentally suckling mice. The finding that HSP70 is associated with the nucleocapsid directly induces HSP70 expression that leads to the association of HSP70 with viral proteins in the brain of protein of HTNV may suggest a functional role of these complexes in the viral life cycles. The potential value of HSP70 HTNV NP complex is to be used as vaccine to prevent and treat hemorrhagic fever with renal syndrome by eliciting the specific cytotoxic T lymphocyte immune response against HTNV.
