摘 要:目的 :探讨γ -IFN是否和冲击波所致肺损伤差异性相关。方法 :BALB/C和C5 7BL/ 6近交系小鼠采用BST -Ⅰ型生物激波管致伤。酶联免疫吸附试验测定血清和肺组织中中γ -IFN的含量 ,Northern杂交试验检测γ -IFN基因在肺组织的表达。结果 :两品系小鼠伤前血清和肺组织γ -IFN含量极低。C5 7BL/ 6小鼠血清标本中γ -IFN含量在伤后 1h显著升高 ,伤后 6h有明显下降。而BALB/C小鼠血清标本γ -IFN含量在伤后 6h显著升高 ,基本与C5 7BL/ 6小鼠伤后 1h水平相当。两种小鼠肺组织γ -IFN含量的变化趋势与血清标本的基本一致 ,不同之处在于伤后 6h ,BALB/C小鼠肺组织γ -IFN含量虽然有显著升高 ,但尚明显低于C5 7BL/ 6小鼠伤后 1h水平 ,而C5 7BL/ 6小鼠γ -IFN含量在伤后 1h和 6h持续维持在一个较高的水平。γ -IFN基因在两品系小鼠中的表达也具有明显的差异性 ,其变化趋势与肺组织中γ -IFN含量的变化相似。结论 :在相同冲击波作用下 ,BALB/C和C5 7BL/ 6小鼠γ -IFN表达呈现显著的差异性 ,提示它可能为这两种小鼠肺损伤差异的原因之一。Objective:To investigate whether there are differences of interferon (IFN) -gamma expression between blast-induced lung injury sensitive (BALB/C) and resistant (C57BL/6) mice.Methods:The BALB/C and C57BL/6 inbred mice were injured by the same blast wave.The expression of IFN-gamma were assayed with Enzyme-linked immunoadsordent assay (ELISA) and Northern blot.Results:The baseline of IFN-gamma productions was very low in both BALB/C and C57BL/6 mice.IFN-gamma production in serum of C57BL/6 elevated significantly at 1 hour,and descended at 6 hours after injury,but in BALB/C mice,it increased only at 6 hours and almost reached the level of that at 1 hour in C57BL/6.The production changes of IFN-gamma in lung tissues of both strains mice also appeared to be different.Unlike those in serum,although the IFN-gamma productions in BALB/C at 6 hours increased significantly,they also were less than those in C57BL/6 at 1 hour.In C57BL/6 mice they maintained a very high level from 1 to 6 hours after injury.The patterns of IFN-gamma gene expression was like those of IFN-gamma productions in lung tissues and there was a significant difference between both strains.Conclusions:Differences in interferon-gamma expression patterns between the strains of mice may contribute to the variable strain susceptibility in blast-induced lung injury.
