摘  要:研究双氢青蒿素、青蒿琥酯体外抗卡氏肺孢子虫 (Pc)不同时相的超微结构改变。将HepG 2细胞接种入若干个 50ml培养瓶 ,待细胞长满瓶底的 80 %时 ,接种Pc。 4h后分别加入双氢青蒿素 5、50 μmol/L ,青蒿琥酯 1 0 0 μmol/L ,喷他脒 (Pentamidine) 1 5μmol/L ,空白对照和 0 3 %无水乙醇。前 3组 (青蒿素衍生物组 )分别于用药后 2、4、8、1 2、1 6、2 4h ,后 3组于用药后 2 4h取样制备电镜标本。结果 用喷他脒处理包膜发生改变。双氢青蒿素 5μmol/L作用 8h后滋养体开始出现改变 ,而 50 μmol/L作用 4h后即有变化 ,其超微结构改变 ,主要为空泡形成和膜系结构的损伤 ,包囊在 2 4h出现病理改变。青蒿琥酯 1 0 0 μmol/L 8h有少量滋养体出现病理变化 ,改变的性质与双氢青蒿素相同。双氢青蒿素在小剂量时 8h ,大剂量时 4h即可对其产生作用 ,主要损害卡氏肺孢子虫的膜系结构。青蒿琥酯的作用较双氢青蒿素弱。To study the ultrastructural changes of Pneumocystis carinii treated with dihydroarteminsinin and sodium artesunate in various times. Transmission electron microscopic observation was done with Pc samples taken at 2, 4, 8, 12, 16 and 24h after treated by dihydroarteminsinin (DHA) at concentratians of 5 and 50 μmol/L and sodium artesunate (ATS) at 100μmol/L. The samples were taken at 24h after treated with pentamidine(PTD)15 μmol/L , 0 3% ethanol and blank control. DHA damaged Pc in low concentration after 8 h and in high concentration after 4h. The ultrastructure of Pc showed damage of cell membrane, vacuoles formation, swollen mitochondrion and endoplasmic reticulum, and broken nuclear membrane. Cysts were damaged at 24h. There were small amount of trophozoites with ATS 100μmol/L damaged at 8h. DHA damaged Pc in low concentration after 8h and in high concentration after 4h. The effect of DHA with different concentrations showed some differences in speed, but no difference in position and character of damages. Trophozoites were more liable to be damaged than cysts. Effect of DHA was stronger than ATS.
