摘 要:目的 探讨一氧化氮 (NO)与白细胞精子症不育的关系。方法 参照WHO标准方法 ,进行精液常规分析。采用过氧化物酶染色法检测精液中白细胞密度。用改良的低渗肿胀法检测精子细胞膜的完整性。采用镀铜镉还原荧光法 ,检测精液中NO代谢产物硝酸盐 (NO 3 )。结果 白细胞精子症不育组白细胞的密度为 (1.4 8± 0 .90 )× 10 9/L ,NO水平为 (10 3.5± 2 .0 ) μmol/L ,显著高于正常生育组的(0 .73± 0 .2 8)× 10 9/L和 (41.6± 1.8) μmol/L (P分别为<0 .0 5和 <0 .0 0 1)。精子的活动率、精子速度试验 (SVT)及精子头、尾部膜完整率 ,均明显低于生育组 (P <0 .0 1) ,而精子的死亡率则显著高于生育组 (P <0 .0 1)。结论 NO水平与白细胞密度呈正相关 ;与精子的活动率、SVT及精子头、尾膜的完整率呈负相关。表明当精液中的白细胞增高时 。Aim To explore the relationship between nitric oxide and leucocyte sperm disease infertility. Methods Conventional sperm analysis was carried out according to WHO standard method. Leucocytic density in semen was detected by peroxidase staining. Integrity of spermatid membrane was examined by improved hyponotic swelling test. A coppered reduced cadmium fluometric assay was used to detect nitrate (NO 3), a metablic product of nitric oxide in semen. Results Semen leucocytic density and nitric oxide level in leucocyte sperm disease infertility group\[(1.48±0.90)×10 9/L and (103.5±2.0) μmol/L, respectively\] were significantly higher than those in normal fertility group \[(0.73±0.28)×10 9/L and (41.6±1.8) μmol/L, respectively\]. ( P <0.05 and P <0.001, respectively). Motile rate of sperm , sperm'velocity test(SVT) and intact rate of sperm head and tail membranes in leucocyte sperm disease group were markedly lower than those in fertility group ( P <0.01), while mortality of sperm was quite the contrary ( P <0.01). Conclusion Nitric oxide level is positively correlated with leucocytic density , wherease negatively correlated with motile rate of sperm, SVT and intact rate of sperm head and tail membranes. It shows that excessive leucocytes in semen may produce excessive nitric oxide which poisons and damages sperm, thus reduceing fertility rate.
