摘 要:通过对86株毛白杨优树和古树的不同组织材料的16种同工酶酶谱分析研究,发现有10种酶系统遗传表达稳定.其中有8种酶系统(ACO,GOT,LAP,NDH,PER,PGM,6-PGDH和SKDH)共有12个基因位点,另外两种酶系统(IDH和MDH)难以确定位点,各具5种酶1谱表现型.用这些酶系统可将86株毛白杨归属于79个不同遗传组成的无性系.重叠率为8.14%,平均重叠率为1.63%.由此证明全国毛白杨档案库收集的资源,具有丰富的种群代表性和可靠性.本文对用同工酶基因标记鉴别无性系所选用酶系统数目及其最佳酶系统组合也进行了讨论.Isozymes inheritance and variation were studied in Chinese white poplar (Populus tomentosa Carr. ) by means of electrophoretic technique. The material tested was sampled from clonal archive of Populus tomentosa in Guanxian County, Shandong Province. All plus trees and old trees in the clonal archive were selected and collected from main distribution zone of Populus tomentosa in China. Isozymes from 16 enzyme systems were extracted from six type organ tissues and separated by horizontal starch-gel eletrophoresis. The enzyme systems were genetically stable including eight systems with 12 loci and two systems with 2×5 isozyme patterns. 86 individual plus trees and old trees were classified under 79 distict clones using sets of combined genetic make-up of the ten enzyme systems.Clone overlap rate(CO) was 8.14% and average clone overlap rate(AO) was 1.63%. It was demonstrated that the rich gene resources were conserved in the clonal archive. The ideal numbers and perfect combination of isozyme systems during the analysis were also discussed.
