摘  要:目的:研究转醛醇酶在不同发育阶段小鼠睾丸中的表达并分离鉴定其亚型。方法:应用SDS-聚丙烯酰胺凝胶电泳(SDS-PAGE)、蛋白免疫印迹(Westernblotting)对转醛醇酶在不同发育阶段小鼠睾丸中的表达进行半定量分析,同时利用双向电泳免疫印迹技术分离、鉴定转醛醇酶的亚型。结果:①转醛醇酶在生后1周小鼠睾丸中表达量平均比成年高47.8%(P<0.005)。②双向电泳Westernblotting示小鼠睾丸转醛醇酶共有10余个亚型,生后1周小鼠睾丸中a8亚型表达量极低,而在成年小鼠睾丸中a8亚型高表达。结论:①生后1周小鼠生长发育快,合成代谢旺盛,转醛醇酶基因绝对表达量高。②a8亚型可能与生殖功能(精子发生、睾酮生成)密切相关。Objective:To study the expression of transaldolase, isolation and identification of its isoforms in different developmental stages of testes in mice. Methods:Transaldoalse expression in different developmental stages of testes in mice were measured by the methods of SDS PAGE and Western blotting, and its isoforms were isolated and identified with two dimensional PAGE Western blotting. Results:①Expression of transaldolase in testes of one week old mice was 47.8%higher than that in adult mice (P<0.005). ②2D PAGE Western blot results showed transaldolase of testes had more than 10 isoforms. The A8 isoform was hardly expressed in one week old mice, while in adult mice it was highly expressed. Conclusion:①The expression level of transaldolase of testes in one week old mice was higher than that in adult mice. ②The a8 isoform of transaldolase was related to spermatogenesis and testosterone synthesis.
