摘　　要：目的　观察IL 2表达质粒对副肌球蛋白 (AgB)核酸疫苗的免疫调节作用。方法　将人的IL 2全长cDNA插入到真核表达质粒pcDNA3中构建成pcDNA3 IL 2重组质粒 ;然后接种C5 7BL 6小鼠 (分为pcDNA3组、AgB组、联合免疫组 ) ,ELISA法检测小鼠血清中的IgG和IgG2a的水平 ;用MTT法检测小鼠脾细胞增殖反应 ,用ELISA法检测其分泌的IL 2和IL 4的水平。最后在仔猪中进行攻击实验 ,观察pcDNA3 IL 2质粒的免疫刺激作用。结果　注射疫苗后 ,pcDNA3 IL 2协同注射组增强了副肌球蛋白特异性的IgG和IgG2a的水平 ,增强了脾细胞的增殖反应 ,提高了其分泌IL 2的水平 ,但对IL 4的水平产生轻微影响。仔猪攻击实验中 ,共同注射IL 2基因组提高了仔猪的相对保护率。结论对于副肌球蛋白核酸疫苗而言 ,人的ILObjective To investigate the immunoregulatory effect of plasmid expressing human interleukin 2 on paramyosin(AgB) DNA vaccine. Methods Recombinant plasmid pcDNA3 IL 2 was constructed by inserting full length human interleukin 2 cDNA into eukaryotic expression vector pcDNA3. C57BL/6 mice were injected intramuscularly with nucleic acid vaccines(including pcDNA3 group, AgB group, combined immunization group). Specific IgG and IgG2a in serum were tested by ELISA. T H cell proliferation and IL 2, IL 4 secreted by splenocytes were determined by MTT method and ELISA kit, respectively. Then challenge trial was performed in newborn pigs. Results Injection of AgB DNA vaccine and IL 2 expressing plasmid combination increased the levels of IgG and IgG2a in mouse serum. T H cell proliferation and interleukin 2 secreted by splenocytes were dramatically increased by co injection of IL 2 gene. However, IL 4 level was influenced insignificantly. Co injection with IL 2 gene increased relative protection rate in newborn pigs. Conclusion Human IL 2 gene is a potent adjuvant for paramyosin nucleic acid vaccine.
