摘  要:目的 :探讨用改进差示分光光度法测定血清苯巴比妥浓度。方法 :在酸性条件下 ,以三氯甲烷二次提取血清中的苯巴比妥 ,利用苯巴比妥在碱性、酸性递质中吸收度差值建立标准曲线 ,按差示分光光度法测定血清中苯巴比妥浓度。结果 :血清苯巴比妥浓度线性范围 2~ 80 μg·mL 1 ,平均回收率 (10 0 .66± 0 .3 6) % ,日内RSD(2 .17± 0 .0 8) % ,日间RSD(2 .5 9± 0 .2 4) % ,血清苯巴比妥在pH =10 .0 0条件下 ,2 5min内较稳定 ,抗干扰性良好。结论 :该方法简便、快捷、灵敏度高、重复性好 ,所需的设备及试剂经济易得 ,适于中小医院血药浓度检测。Objective:To improve differential spectrophotometry for the determination of phenobarbital in serum. Method:Phenobarbital in serum was twice extracted with trichoromethane under acidic conditions. The standard curve of phenobarbital in serum was established according to the absorbance deviations of phenobarbital in base and acidic media, and the phenobarbital in serum was determined by differential spectrophotometry. Results:The linear range of phenobarbital in serum was 2~80 μg·mL -1 , the average recovery was (100.66±0.36)% with RSD (2.17±0.08)% and (2.59+0.24)% for intraday and interday respectively. The phenobarbital in serum was stable in 25 min under base conditions of pH=10.00 Conclusion:The method now used is quick, simple, of high sensitivity and of good repeat. It is cheap and easy to obtain the instrument and reagent and suitable for clinical drug monitoring in small and medium-sized hospitals.
