摘  要:目的 了解海南省恶性疟原虫裂殖子表面蛋白 1(pfMSP1)和表面蛋白 2 (pfMSP2 )等位基因型的分型特征。方法 采用套式PCR法分别扩增pfMSP1基因第 2区与第 3区部分片段和pfMSP2基因的中间可变区 (第 3区 ) ,对海南省恶性疟原虫分离株进行基因分型。结果  5 5份血样中有 5 2份恶性疟疾患者扩增出pfMSP1基因片段 ,以MAD2 0型为主导型 (84 6 % ) ,K1型为次要型 ,未检出RO33型 ,两种不同等位基因混合感染率为 15 4 %。同时 ,5 5份血样中有 5 3份恶性疟疾患者扩增出pfMSP2基因片段 ,以 3D7型为主导型 (83% ) ,FC2 7型为次要型 ,混合感染率为 17%。结论 海南省恶性疟原虫的MSP1存在MAD2 0型和K1型两种等位基因型 ,以MAD2 0型为优势虫株 ;其MSP2存在 3D7型和FC2 7型两种等位基因型 ,以 3D7等位基因家族为主。Objective To identify the genotypes of merozoite surface protein 1(PfMSP1) and surface protein 2(PfMSP2) ofPlasmodium falciparum isolates from Hainan province. Methods Nested PCR was applied to amplify the Block 2 and 3 of PfMSP1 and the variable regions (Block3) of PfMSP2. Results From 52 out of 55 blood samples fromPlasmodium falciparum patients, gene fragments of PfMSP1 were amplified. The majority allele was the MAD20-type allele(84 6%), while the minority allele was the K1-type allele, and no RO33-type allele was found. 53 PfMSP2 gene fragments were amplified from 55 blood samples. 3D7-type was the major alleles(83%) while the minority was the FC27-type allele. The mixed infection rate of two different genotypes with two different alleles of PfMSP1 and PfMSP2 was 15 4% and 17%, respectively. Conclusion Two allelic types of MSP1 gene (MAD20-type and K1-type) and two allelic types of MSP2 gene (3D7-type and FC27-type) existed in malaria endemic areas in Hainan province. MAD20-type and 3D7-type were the major alleles in the endemic area.
