摘 要:目的:研究Leydig细胞在抗感染中的免疫调节作用。方法:SD大鼠睾丸经Ⅱ型胶原酶消化、过滤及Percoll分离获得高纯度、高活率的Leydig细胞。体外培养的Leydig细胞经解脲脲原体(UU)感染后,观察其细胞培养上清液中白介素-I(interleukin-1,IL-1)活性(四唑盐比色法即MTT法)和转移生长因子(transforming growth factor-β_1,TGF-β_1)含量(ELISA法)的变化。结果:UU感染可明显促进Leydig细胞分泌IL-1和TGF-β_1的能力,且与UU的感染剂量有关。结论:大鼠Leydig细胞在抗感染免疫中,可通过其分泌的IL-1和TGF-β_1发挥免疫调节作用。Objective:To investigate immune regulation of the rat Leydig cells in the infection. Methods- The testis of the SD rat was digested with collagenase (type II) and after filtraing and isolation by Percoll, the Leydig cells (higher purifity)were obtained. After the Leydig cells, were infected by Ureaplasma ure-aticum (UU) in vitro, the effect of UU infection on the IL-1 and TGF-β1 secretion by rat Leydig cells was analysed by MTT and ELISA respectively. Results: The IL-1 activity and TGF-β1 level were significantly increased, which secreted by rat Leydig cells in the UU infection. Conclusion: Leydig cells regulation through the secretion of the IL-1 and TGF-β1 in the infection of the testis by UU in rat.
