摘 要:目的 探讨外周血淋巴细胞 (peripheralbloodlymphocyte ,PBLs)中穿孔素 (perforin ,P)和颗粒酶B(granzymeB ,GB)在肾移植抗急性排斥反应时的表达。方法 利用定量RT PCR方法动态观察了 10例肾移植患者发生急性排斥反应后抗排斥治疗时P和GB的表达。结果 10例肾移植患者抗排斥治疗后 ,PBLs中P和GB表达水平明显下降。结论 利用定量RTObjective To investigate the expression of perforin(P) and granzyme B (GB) of peripheral blood lymphocyte (PBLs) during antirejection therapy of human renal allografts. Methods We used quantitative reverse transcription polymerase chain reaction(RT PCR) to dynamic analyze the expression of P and GB in PBLs from 10 renal allograft recipients during antirejection therapy, compared with the level of serum creatinine.Results During antirejection therapy of 10 renal allograft recipients, the level of expression of P and GB in PBLs was obviously downregulated. Their down tendency was ahead of serum creatinine.Conclusion Using quantitative RT PCR to dynamic analyze the expression of P and GB in PBLs may serve as a noninvasive, sensitive method to monitor the effect of antirejection therapy.
