摘  要:1985年从新疆石河子地区糖用甜菜上分离到一个病毒分离物,样品代号85-04。生物测定证明其寄主范围主要限于藜科。可系统侵染甜菜、菠菜引起系统退绿斑驳花叶;系统侵染苋色藜、昆诺阿藜和番杏,引起黄褐色坏死斑。病毒的体外热钝化温度为55~66℃。体外存活期1~2天,稀释限点1/4000。可由桃蚜等多种蚜虫进行非持久性传播。用PEG沉淀,硫酸铯平衡密度梯度离心,可获得较纯的病毒。病毒形态为线状,长约730nm,直径13nm。病叶的超薄切片中可见到大量的风轮状,旋涡状和束状内含体。用纯化的病毒制备出效价为32的抗血清,能与甜菜花叶病毒产生明显的沉淀线。由以上研究结果证实85-04分离物是甜菜花叶病毒(Beet Mosaic Virus,BtMV)。用聚丙烯酰胺凝胶电泳测出该病毒的外壳蛋白有两条区带、分子量分别为35075和25118道尔顿,前者为单一亚基病毒外壳蛋白,该蛋白由17种氨基酸组成(由于色氨酸在水解过程中被破坏,未能测定)25k区带为降解产物。An isolate, No.85-04, was collected from infected surgarbeet with chlorosis and mottling at Shihezi Xingiang in 1985 and biologically purified through several local lesion passages. Its host range is very narrow, only limited to Chenopodiaceae. It causes systemic yellow brown spots in Chenopodiaceae, C.quinoa and Tetra9onia expansa,and a systemic chlorotic mottle in sugar beet and spinach. Its dilution end-point was found to be 1 : 5,000. The virus is activated by exposure to temperature between 55-60C for 10 min. The longevity in vitro of the crud sap from infected beet leaves is between 24-43 hrs.at room temperature. The virus is sap-inoculable and transmitted by aphids,especially Myzus per sicae. The purified virion is filamentous about 730nm long and 13nm in width.Ultrathin section of the infected or leaves show various inclusions in the cytoplasm,lither pinwheel or bundle or scrollshaped. The antiserum against the virus gave a single precipitation line with the type culture of BtMV identifuied by Beijing Agricultural University .The isolate is identified as Beet Mosaic Virus based on the results mentioned. Two bands, -35k and-32k, of the dissociated virus particles were detected in a 10% SDS-PAGE. The smaller one appear to be a degradation product. The amino acid composition of the viral coat protein was also analyzed.
