补肾益气活血方对宫内发育迟缓胎鼠脑细胞内游离钙离子浓度的影响

摘　　要：探讨补肾益气活血方对宫内发育迟缓 (interuterine growth retardation,IU GR)胎鼠脑细胞内游离钙离子浓度的影响。采用被动吸烟法建立缺血缺氧 IUGR大鼠模型 ,将动物分成 IUGR模型组 (IU GR组 ) ,IU GR中药治疗组(中药组 ) ,IUGR L - argine治疗组 (精氨酸组 )和正常对照组 (正常组 )。运用细胞内 Ca2 + 荧光指示剂 Fura- 2 /AM检测各组中妊娠第 18～ 2 1d胎鼠神经元突触体胞液游离钙离子浓度 ,并以胎鼠出生体重和脑重作疗效评价。结果 IUGR组胎鼠体重和脑重较之正常组明显降低 (P<0 .0 1) ,中药组和精氨酸组胎鼠体重和脑重较之 IUGR组明显升高 (P<0 .0 1) ,且中药组胎鼠体重和脑重已接近正常组 (P>0 .0 5 )。自妊娠第 18d可见 IUGR组胎鼠脑细胞内游离 [Ca2 + ]i已开始升高 ,至分娩前 (第 2 1d)为正常组值的 3倍多 ,中药组与正常组 [Ca2 + ]i相对稳定 ,而精氨酸组有轻微上升。说明补肾益气活血方能通过抑制钙离子超载而保护胎鼠脑细胞 。To investigate the effects of the prescription of reinforcing kidney, replenishing Qi and activating blood (PRKRQAB) on the cerebral cytoplasmic free calcium concentration of fetal rats with fetal growth retardation (IUGR) , the passive smoking was used to establish hypoxic ischemic injury model of IUGR rats. The rats were divided into four groups: normal group, IUGR group, L argine group and Chinese herb medicine group (CHM group). Intracellular Ca 2+ fluorescence luminal reagent Fura 2/AM was used to measure the \[Ca 2+ \] i of cerebral neuronal synaptosome of fetal rats from day 18 to 21 during pregnancy in each group. The mean fetal birth weight and brain weight were also observed as an evaluation criterion of effects. Results revealed that the mean fetal birth weight and brain weight in IUGR group were decreased greatly as compared with those in the normal group ( P <0 01). Compared with IUGR group, the fetal birth weight and brain weight were increased significantly both in CHM group and L argine group ( P <0 01), but there was no significant difference between CHM group and normal group ( P >0 05). From pregnant day 18, \[Ca 2+ \] i of IUGR group began to increace and was more than 3 times before labor (pregnant day 21) as the normal group at the same time. \[Ca 2+ \] i of CHM group was always stable but slightly increaced in L argine group. It was demonstrated that the PRKRQAB can protect neural cells of IUGR fetal rats through inhibiting intrasynaptosomal calcium overload. The effects of PRKRQAB were more satisfactory than L argine.
