摘　　要：在人类基因组完成工作框架图以后 ,人类功能基因组学成为最重要的任务之一。我们目前正在重点研究编码细胞因子和凋亡相关分子的人类功能基因。本综述介绍其中的 2个首先由我们实验室克隆的分子 ,即趋化素样因子 1(CKLF1)和程序化细胞死亡分子 5 (PDCD5 ,过去命名为TFAR19)。CKLF1是属于一个新的基因超家族的成员 ,具有体内外对不同白细胞的趋化活性、刺激骨骼肌增殖活性和刺激骨髓细胞增殖活性。在临床应用上 ,重组CKLF1蛋白质有可能用于肌萎缩的治疗 ,CKLF1抑制剂有可能用于变态反应和自身免疫病的治疗。PDCD5是凋亡的正调节分子 ,能够结合胱天蛋白酶 3,并在体外延长其半衰期。较多证据表明 ,PDCD5在肿瘤组织的表达低于正常组织。PDCD5有可能成为肿瘤。After the human genome project (HGP) was complete d, one of the most import ant tasks is to explore human functional genomics. We are now just focusing on t he discovery and functional clarification of novel human genes encoding cytokine s and apoptosis-related molecules. This review will give a brief introduction t o two of the molecules, namely chemokine like factor 1(CKLF1) and programmed cel l deaths 5( PDCD5 , formerly named TFAR19 ), both of which were firstly clo ned from our laboratory. The CKLF1, which belongs to a novel gene family, has chemotaxis effect on differ ent leukocytes both in vitro and in vivo . It can also stimulate prolifer ation of skeletal muscle cells and bone marrow cells. The recombinant CKLF1 may have app lication value in the treatment of muscle atrophy, and CKLF1 inhibitors could be used against allergy autoimmune diseases.The PDCD 5 is a positive regulator for the apoptosis. It can bind caspase-3 and p rolong its half-life in vitro . Multiple evidences indicate that the expres sion of PDCD 5 is lower in tumor tissues than that in normal tissues. The PDCD 5 could be used as a mark or target for some apoptosis related diseases, such as tumor a nd autoimmune diseases.
