摘　　要：为建立大肠杆菌细菌脂多糖(LPS)体外诱导牦牛子宫内膜细胞炎症的模型,在生长良好的牦牛子宫内膜上皮细胞中分别加入浓度为1、50、100、200、500、1000 ng/mL的LPS,采用MTT法检测其细胞毒作用,确定LPS最佳浓度;在牦牛子宫内膜上皮细胞中添加最佳浓度LPS,孵育至24、48、72 h,分别收集培养上清液,采用ELISA法测定白细胞介素1β(IL-1β)和肿瘤坏死因子-α(TNF-α)分泌变化情况。结果表明,LPS体外诱导牦牛子宫内膜炎症模型的最佳浓度为100 ng/mL,在孵育至24 h及以上时,IL-1β和TNF-α的质量浓度与对照组相比差异显著(P<0.05),表明该炎症模型建立成功。In order to establish an in vitro model of yak endometrial cell inflammation induced by escherichia coli lipopolysaccharide,well-grown yak endometrial epithelial cells were respectively injected with LPS concentrations of 1,50,100,200,500 and 1000ng/mL to detect the cytotoxicity by MTT and determine the optimum concentration.Then,the optimum concentration of LPS was injected to yak endometrial epithelial cells and was incubated for 24,48 and 72 Hrs.The culture supernatant was respectively collected to determine the secretion variation of interleukin 1β(IL-1β)and tumor necrosis factor-α(TNF-α)by ELISA.The results showed that the optimum concentration of LPS in vitro inducing yak endometritis model was 100ng/mL,and the mass concentrations of IL-1βand TNF-αwere significantly different from those of the control group after incubating for 24 Hrs or more(P<0.05),which indicated that the inflammatory model was successfully established.
