摘 要:目的探讨葛根素对绝经后骨质疏松大鼠的护骨素、核因子-κB受体活化因子配体、碱性磷酸酶和骨组织的影响。方法5月龄健康SD雌性大鼠96只,随机分为:假去卵巢(Sham)组;绝经后骨质疏松模型(OVX)组、山茶籽治疗(Ts)组、雌二醇(E2)治疗(E2)组。比较各组第2、6、8和12 w,护骨素、核因子-κB受体活化因子配体和碱性磷酸酶水平,观察各组股骨头组织结构。结果OVX组股骨头组织有明显疏松病理改变,护骨素明显低于Sham组、Ts组和E2组。OVX组核因子-κB受体活化因子配体、血清碱性磷酸酶明显高于Sham组、Ts组和E2组。结论葛根素有雌激素样作用对绝经后骨质疏松有防治作用。Objective Understanding of puerarin on postmenopausal osteoporosis model rats bone element,nuclear factor kappa B receptors predominate activation factor ligands,alkaline phosphatase and bone tissue and the effect of puerarin forⅠtype and provide a referrence for the prevention and treatment of osteoporosis.Methods 96 healthy SD female rats aged 5 months were randomly divided into 4 experimental groups:sham ovariectomized group(sham);Postmenopausal osteoporosis model(OVX);Camellia seed treatment group(Ts);Estradiol treatment group(E2).At week 2,6,8 and 12,6 rats in each group were randomly selected to take 5ml blood from the anaesthetic heart cavity and then sacrificed.The proximal left femoral head was taken for 1cm section to observe the morphological structure.Serum was obtained for the determination of osteoctin,nuclear factor-B receptor activator ligand and alkaline phosphatase.Results The 1cm bone tissue of the femoral head in the postmenopausal osteoporosis model rats showed obvious pathological changes of osteoporosis,and the serum osteophytin of the OVX group was significantly lower than that of the Sham group,Ts group and E2 group.However,the ligands of nuclear factor-kappa B receptor activator and serum alkaline phosphatase were significantly higher in OVX group than in Sham group,Ts group and E2 group.Microscopically,osteoporosis was significantly improved in the two drug interference groups There was no significant difference in bone tissue,serum osteocalcin,nuclear factor-kappa B receptor activator ligand and serum alkaline phosphatase between the two treatment groups and Sham group.Conclusions Puerarin has estrogen-like effects;Puerarin can improve the osteocalcin,decrease the action of nuclear factor-kappa B receptor activating factor ligand and serum alkaline phosphatase in postmenopausal osteoporosis model rats.
