摘 要:目的探究芒柄花黄素对宫颈癌细胞放疗敏感性的影响。方法芒柄花黄素联合放疗处理宫颈癌HeLa细胞为芒柄花黄素联合放疗组,单独放疗处理HeLa细胞为对照组,利用噻唑蓝(MTT)法和克隆形成实验分别测定两组细胞的增殖抑制率和克隆形成能力,流式细胞仪和Western印迹分别检测细胞的凋亡率和细胞增殖相关蛋白Ki-67的表达情况。结果芒柄花黄素联合放疗组细胞增殖抑制率、凋亡率和细胞增殖相关蛋白Ki-67的表达量均明显高于对照组(P<0.05),克隆形成率明显低于对照组(P<0.05)。结论芒柄花黄素可以明显提高宫颈癌细胞的放疗敏感性,为芒柄花黄素应用于宫颈癌的放疗治疗提供了理论依据。Objective To investigate the effect of formononetin on radiosensitivity of cervical cancer cells.Methods After HeLa cells were treated with formononetin and radiation,MTT method and colony formation assay were used to assess cell proliferation inhibition rates and colony formation ability of cervical cancer cells.Flow cytometry and Western blot were used to detect apoptosis rates and expression level of proliferation-associated protein Ki-67 of HeLa cells.Results After treatment with formononetin and radiation,the cell proliferation inhibition rates,apoptosis rates and expression level of proliferation-associated protein Ki-67 of HeLa cells were all higher than those in control group(radiation treatment only),while the colony formation ability was significantly decreased compared with that in control group.Conclusions Formononetin could dramatically improve the radiosensitivity of cervical cancer cells,which provid a theoretical basis for the application of formononetin in radiotherapy of cervical cancer.
