摘 要:目的:研究4-羟基-2,2,6,6-四甲基哌啶(tempol)对糖尿病大鼠肾脏和脊髓p22^phox的影响。方法:选取雄性SD大鼠造模,糖尿病大鼠模型造模成功后,随机分为糖尿病组和干预组,另设正常对照组进行实验,干预组tempol灌胃,其他组等量生理盐水灌胃。4 w后称重麻醉处死,迅速取肾、脊髓脱水固定,冰冻切片,NADPH氧化酶亚基p22^phox免疫组化染色,数码显微拍照判读分析。结果:免疫组化染色后,肾脏和脊髓细胞膜上p22^phox颜色染为棕黄色圆圈或线条,对照组呈阴性极少见分布,糖尿病组呈强阳性弥漫性分布,干预组呈现弱阳性分散性分布。3组之间的阳性细胞比较,差异有统计学意义(P<0.05)。结论:用Tempol干预糖尿病大鼠,可抑制p22^phox的生成,使肾脏和脊髓氧化应激水平降低,对糖尿病大鼠有一定的保护作用。Objective:To study the effects of 4-hydroxy-2,2,6,6-tetramethylpiperidine(tempol)on p22^phox in kidney and spinal cord of diabetic rats.Method:Male SD rats were selected to establish the model.After successful establishment of the model,the diabetic rats were randomly divided into diabetic group and intervention group.Another normal control group was set up to carry out the experiment.The intervention group was administered tempol,and the other groups were administered the same amount of saline.Four weeks later,the kidneys and spinal cord were dehydrated and fixed,frozen sections,immunohistochemical staining of NADPH oxidase subunit p22^phox,and interpretation analysis of digital microphotography were performed.Result:After immunohistochemical staining,the p22^phox color on the membranes of kidney and spinal cord cells was brown and yellow circles or lines.The negative distribution was rare in the control group,the strong positive diffuse distribution was found in the diabetic group,and the weak positive and dispersive distribution was found in the intervention group.There was significant difference in positive cells among the three groups(P<0.05).Conclusion:Tempol can inhibit the production of p22^phox in diabetic rats,reduce oxidative stress in kidney and spinal cord,and protect diabetic rats.
