生骨密骨方对去卵巢骨质疏松大鼠血清骨代谢标志物的影响

摘　　要：目的:研究分析生骨密骨方对卵巢去势(OVX)骨质疏松(OP)大鼠骨血清骨代谢标志物中骨特异性碱性磷酸酶(B-ALP)、骨钙素(BGP)、I型胶原羧基端肽β降解产物(β-CTx)、血清抗酒石酸酸性磷酸酶(TPACP5b)的影响。方法:选取48只6月龄雌性未生育健康SD大鼠,手术摘除其双侧卵巢,建立骨质疏松大鼠动物模型。建模后随机分为3组各16只:对照组(OV组)、雌二醇组(OE组)、生骨密骨组(OT组)。术后OV组灌服生理盐水,OE组给予戊酸雌二醇片,OT组给予生骨密骨方水煎服浓缩液,3组等量给予连续12周,收集血浆,检测血清标志物B-ALP、β-CTx、BGP和TPACP5b,比较3组之间的结果。结果:与OV组相比较,OT组及OE组大鼠血清B-ALP、β-CTx、TPACP5b浓度水平降低,BGP水平显著增加(P<0.05),且OT组与OE组在大鼠血清B-ALP、β-CTx、TPACP5b及BGP标志物浓度水平的差异无显著性(P>0.05)。结论:生骨密骨方能够通过改变骨代谢标志物中的浓度水平,起到防治骨质疏松的疗效且效果显著。Objective:To study the effects of Shenggu Migu Recipe on bone specific alkaline phosphatase(B-ALP),osteocalcin(BGP),collagen type I carboxyl terminal peptide beta degradation product(β-CTx)and serum tartrate-resistant acid phosphatase(TPACP5b)in ovariectomized(OVX)rats with osteoporosis(OP).Methods:Forty-eight healthy female SD rats aged 6 months were selected and their bilateral ovaries were removed surgically to establish an osteoporotic rat model.Then they were randomly divided into three groups:control group(OV group),estradiol group(OE group),Shenggu Migu group(OT group).After operation,OV group was given normal saline,OE group was given estradiol valerate tablets,OT group was given Shenggu Migu Decoction concentrate,and the three groups were given the same amount for 12 weeks.Plasma was collected,B-ALP,β-CTx,BGP and TPACP5b were detected,and the results between the three groups were compared.Results:Compared with OV group,B-ALP,β-CTx,TPACP5b levels in OT group and OE group decreased,and BGP levels increased significantly(P<0.05).There was no significant difference in serum B-ALP,β-CTx,TPACP5b and BGP markers between OT group and OE group(P>0.05).Conclusion:Shenggu Migu Recipe can prevent and treat osteoporosis by changing the concentration of bone metabolic markers.
