摘 要:采用实时荧光定量PCR技术,分别对3、7、12和16周龄拜城油鸡肌肉组织中脂肪细胞型脂肪酸结合蛋白(A-FABP)、心型脂肪酸结合蛋白(H-FABP)和过氧化物酶体增殖物激活受体(PPAR-γ)基因mRNA表达量进行分析,结合肌内脂肪(IMF)含量测定,分析H-FABP、A-FABP和PPAR-γmRNA表达的发育性变化及其与IMF的相关性。结果显示:拜城油鸡肌肉组织中A-FABP mRNA相对表达量随周龄的增长呈上升趋势,其中A-FABP基因在胸肌16周龄时的相对表达量显著高于3、7和12周龄(P<0.05),该基因在12、16周龄时腿肌中的表达量显著高于3和7周龄(P<0.05);H-FABP mRNA表达量随着拜城油鸡周龄增加呈现下降趋势,但各周龄间均不存在明显的发育性变化(P>0.05);PPAR-γ基因在公鸡胸肌16周龄时的mRNA表达量最高,且显著高于3、7和12周龄(P<0.05),公、母鸡腿肌PPAR-γmRNA表达量随着周龄呈逐渐上升趋势,在16周龄时的表达量最高,且显著高于3周龄(P<0.05)。不同发育阶段拜城油鸡肌肉组织中A-FABP、H-FABP和PPAR-γmRNA表达量与其IMF含量呈不同程度的相关性,A-FABP mRNA表达量与其IMF含量呈现极显著的正相关(P<0.01);H-FABP mRNA表达量与IMF含量存在负相关,但相关性均未达到显著水平(P>0.05);PPAR-γmRNA表达量与IMF含量间存在正相关,但相关性均未达到显著水平(P>0.05)。由此推测,A-FAPB基因对IMF的沉积具有调控作用,并可作为拜城油鸡IMF性状进行分子标记辅助选育的遗传标记之一。The mRNA expression levels of adipocyte fatty acid-binding protein(A-FABP), heart fatty acid-binding protein(H-FABP), and peroxisome proliferator-activated receptor gamma(PPAR-γ) genes in muscle tissues at different developmental stages(3, 7, 12 and 16 weeks of age) of Baicheng You chicken were detected by real-time fluorescence quantitative PCR, and measured the intramuscular fat(IMF) contents in muscle tissues, analyzed the correlations between genes mRNA expression level and IMF. The results showed that the relative mRNA expression of A-FABP gene in the muscle tissue of Baicheng You chicken showed an increasing trend with the increase of age, the relative expression of A-FABP gene at 16 weeks of age was significantly higher than those of 3, 7 and 12 weeks of age(P<0.05), the expression of this gene in leg muscle at the age of 12 and 16 weeks was significantly higher than that at 3 and 7 weeks of age(P<0.05). The mRNA expression of H-FABP decreased with the increase of weekly age of Baicheng You chicken, but there was no difference(P>0.05). The mRNA expression level of PPAR-γ gene was the highest at 16 weeks of age of male chicken breast muscle and significantly higher than those of 3, 7 and 12 weeks of age(P<0.05), the mRNA expression level of PPAR-γ gene of male and hen leg muscle was gradually increased with aging, and was the highest at 16 weeks of age and significantly higher than 3 weeks of age(P<0.05). The mRNA expression levels of A-FABP, H-FABP and PPAR-γ genes in the muscle tissue of Baicheng You chicken during different development stages showed different correlations with the IMF content, and the mRNA expression levels of A-FABP gene presented a very significant positive correlation with the IMF content(P<0.01), the mRNA expression of H-FABP gene was negative but not significant correlated with its IMF content(P>0.05), there was a positive but not significant correlation between the mRNA expression level of PPAR-γ and IMF content(P>0.05).
