摘  要:目的:观察验证PI3K/Akt信号通路对头部电针调控脑瘫鼠脑皮质神经生长相关蛋白水平的影响。方法:设正常组、模型组、头部电针组(干预1组)、头部电针+LY294002组(干预2组)、头部电针+LiCl组(干预3组);构建缺血缺氧性脑瘫大鼠模型,以电针刺激头部运动区的头部电针疗法为干预手段,运用Western Blot法检测大脑皮质PI3K/Akt信号通路磷酸化水平,神经生长相关蛋白神经特异性烯醇化酶(NSE)、微管结合蛋白(MAP-2)、髓磷脂碱性蛋白(BMP)表达水平,并结合病理学检查及神经行为学评估结果进行关联分析。结果:模型组、干预2组各时段的运动功能评分明显低于干预1组、干预3组(P<0.05),干预2组PI3K/Akt信号通路磷酸化水平明显低于模型组、干预1组及干预3组(P<0.05);与干预2组比较,干预1组、干预3组的NSE表达下降,MAP-2、MBP表达增高,脑皮质组织病理改变恢复明显。结论:PI3K/Akt信号通路参与电针刺激头部运动区对脑瘫鼠脑损伤区神经生长相关蛋白的调控,在促进神经功能康复的作用机制中发挥重要作用。Objective: To observe and verify the regulation effect of the PI3 K/AKT signaling pathway on nerve growthassociated proteins level in cerebral palsy rats cortex with the intervention of electroacupuncture. Methods: The cerebral palsy rat model of hypoxic-ischemic was constructed with intervention by electroacupuncture at the head, which was divided into normal group, model group, electroacupuncture group(intervention group 1), electoacupuncture+LY294002 group(intervention group 2),electoacupuncture+Li Cl group(intervention group 3). Western Blot was used to detect the phosphorylation of PI3 K/Akt signaling pathway, and the expression level the nerve growth-associated proteins(NSE), microtubule(MAP-2), myelin basic protein(BMP)in the cerebral cortex of each group, which were conducted correlation analysis combined with histopathology examination and neurobehavioral assessment. Results: The motor function scores of PI3 K/Akt signal pathway in model group and intervention group 2 were significantly lower than those in intervention group 1 and intervention group 3(P<0.05), the phosphorylation level of PI3 K/Akt signal pathway in intervention group 2 were significantly lower than those in model group and intervention group 1 and intervetion group 3(P<0.05). Compared with intervention group 2, NSE expression in intervention group 1 and intervention group 3 decreased,MAP-2 and MBP expression increased, and pathological changes in cerebral cortex tissue recovered significantly. Conclusion: PI3 K/Akt signaling pathway participates in the regulation of nerve growth-related proteins in brain injury area of cerebral palsy rats by electroacupuncture stimulation of head motor area, and plays an important role in promoting the functional recovery of nerve.
