摘 要:目的:探讨凉性中药薏苡仁对虚热证模型大鼠能量代谢及其相关基因表达的影响。以能量代谢变化为切入点立足基因,阐释薏苡仁治疗虚热证的分子机制。方法:中药复方(熟附子、肉桂、干姜)灌胃建立虚热证大鼠模型。对温度、基础代谢、体质量、寒热趋向、自主活动及RNA-seq转录组测序检测大鼠能量代谢变化。结果:薏苡仁干预后,自主活动、体质量、基础代谢、体温(红外成像)明显降低(P<0.05)。测序结果显示,共筛出59条DEGs,与代谢直接相关基因共26条。糖代谢(9)、脂代谢(11)、氨基酸代谢(6)。结论:薏苡仁通过调整代谢紊乱治疗虚热证,同时也是薏苡仁清热利湿之功得以发挥的主要分子机制。Objective:To investigate the effects of cool Chinese herbal medicine coix seed on energy metabolism and related gene expression in rats with deficiency heat syndrome.Based on the change of energy metabolism as the entry point,the molecular mechanism of the treatment of deficiency heat syndrome was discussed.Methods:A rat model of deficiency heat syndrome was established by intragastric administration of traditional Chinese medicine compound(cooked aconite,cinnamon and dried ginger).Temperature,basal metabolism,body weight,cold and heat trend and autonomic activity and RNA-seq transcriptome sequencing were used to detect changes in energy metabolism in rats.Results:After treatment with coix seed decoction,the autonomic activity,body weight,basal metabolism and temperature were significantly lower(P<0.05).The sequencing results showed that 59 DEGs in the treatment group,and the functional classification annotation(GO)was significantly changed.A total of 26 DEGs directly related to metabolism.Focus on sugar metabolism(9),lipid metabolism(11),and amino acid metabolism(6).Conclusion:Coix seed can treat deficient heat syndrome by adjusting metabolic disorders.Meanwhile,it is also the main molecular mechanism of the clearing heat and dampness of semen coicis.
