脐疗对寒凝血瘀型痛经大鼠扭体反应及神阙穴区TLR4的影响

摘　　要：目的:观察脐疗对寒凝血瘀型痛经大鼠扭体反应及神阙穴区Toll样受体4(TLR4)表达的影响,探索脐疗效应与神阙穴区的关系及启动机制。方法:大鼠随机分为正常对照组、模型组、药物贴脐组、隔药灸脐组、艾条灸脐组。除正常对照组外,其余各组构建寒凝血瘀痛经模型,给予相应的治疗。记录各组大鼠扭体反应情况,并通过免疫荧光、实时荧光定量PCR技术检测大鼠神阙穴区TLR4表达。结果:与模型组比较,各脐疗组扭体反应缓解(P<0.05,P<0.01);隔药灸脐组对扭体次数及扭体评分的缓解优于药物贴脐组和艾条灸脐组(P<0.05,P<0.01);各脐疗组TLR4 mRNA较模型组明显增加(P<0.01),隔药灸脐组与艾条灸脐组优于药物贴脐组(P<0.01)。结论:3种脐疗方法均能减缓痛经大鼠扭体反应,隔药灸脐法作用最优;3种脐疗方法均能上调寒凝血瘀型痛经大鼠神阙穴区TLR4的表达以激活腧穴功能。Objective: To observe the effects of navel therapy on the expression of TLR4 in Shenque of coagulated cold and blood stasis type dysmenorrhea model rats, to explore the initiation mechanism and the relationship between the umbilical effect and Shenque. Methods: Rats were randomly divided into normal control group(group A), model group(group B), drug sticking navel group(group C), Herb-partitioned moxibustion on navel group(group D), moxa moxibustion navel group(group E). In addition to group A, constructed coagulated cold and blood stasis dysmenorrhea model was constructed in the other groups,and acceptted the corresponding treatment. The writhing response of each group of rats was recorded, immunofluorescence and Quantitative Real-time PCR were used to detect the expression of TLR4 in Shenque. Results: Compared with group B,the writhing response of each navel group was alleviated(P<0.05, P<0.01). The group D was superior to group C and group E in relieving twisting response(P<0.05, P<0.01). The expression of TLR4 mRNA in each navel group were significantly higher than group B(P<0.01), group D and group E were superior to group C(P<0.01). Conclusion: The three kinds of navel therapy all can relieve the writhing response, the therapeutic advantages of herb-partitioned moxibustion on navel therapy is the best. Navel therapy can upregulate the expression of TLR4 in Shenque of coagulated Cold and Blood Stasis type dysmenorrhoea rat to activate the acupoint function.
