摘 要:目的:建立微波消解-电感耦合等离子体质谱(ICP-MS)法测定丹参根茎中30种微量元素的含量,为丹参的生物活性和药理作用的进一步研究提供参考。方法:取3批丹参样品,采用HNO3-H2O2体系将丹参粉末湿法消解,以In、Rh为内标元素,用ICP-MS法测定丹参根茎中微量元素的含量。结果:各元素线性关系良好,r≥0.9996;检测限在0.0005~0.1600μg/L;精密度、稳定性和重复性试验的RSD分别小于4.7%、4.3%和4.9%,符合质量分析要求;加样回收率为90.0%~105.9%,RSD 0.1%~4.3%。其中,硼(B)、钒(V)、铬(Cr)、锰(Mn)、钴(Co)、镍(Ni)、铜(Cu)、锌(Zn)、硒(Se)、铷(Rb)、锶(Sr)、钼(Mo)、锡(Sn)等人体必需微量元素含量较高,铅(Pb)、砷(As)、镉(Cd)、汞(Hg)等重金属元素含量较低,低于《中华人民共和国药典》2015年版的限度要求。结论:该方法快速准确测定了丹参根茎中30种微量元素的含量。通过对微量元素的分析,可以为丹参药材的质量控制、安全性评价提供一定参考。Objective: To establish an inductively coupled plasma spectrometry(ICP-MS) with microwave digestion method for the determination of 30 kinds of trace elements in Radix Salviae Miltiorrhizae, to fill the blank of trace elements of this plant and provide references for further study of the biological activity and pharmacological action of Radix Salviae Miltiorrhizae.Methods: Three batches Radix Salviae Miltiorrhizae were used as samples. The contents of elements were determined by ICPMS after the samples were diluted with wet digested in HNO3-H2 O2 system, then with In, Rh as the internal standard, the contents of elements were calculated with standard curve method. Results: The 30 trace elements showed good linearity in the selected concentration ranges, with the correlation coefficients over 0.9996. The detection limits of the 48 elements were in the range of0.0005~0.1600μg/L. The RSD values of precision, stability, and repeatability were less than 4.7%, 4.3% and 4.9%, respectively,all of which met the demands of quantitative analysis. The recovery was 90.0%~105.9%, while the RSD was 0.1%~4.3%. The content of the essential trace elements such as B, V, Cr, Mn, Co, Ni, Cu, Zn, Se, Rb, Sr, Mo, Sn was high. The content of the heavy metal elements including Pb, As, Cd, Hg was under the limit requirements of People’s Republic of China Pharmacopoeia(2015). Conclusion: The method accurately and quickly determined the content of 30 trace elements in the root of Radix Salviae Miltiorrhizae. The analysis of trace elements can provide certain references for the quality control, safety evaluation, and clinical application of Radix Salviae Miltiorrhizae.
