摘  要:目的:筛选黄硝创伤方抑菌的主要药味,分析并优化最佳配伍。方法:以体外抑菌作用为指标,采用正交t值法,按正交t值表安排的复方药味,制备水煎液12组,采用药敏片法进行体外抑菌实验,通过观察药敏片周围是否出现抑菌圈及其直径大小,测定处方各组对金黄色葡萄球菌标准菌株、临床耐药肠球菌的抑菌效果,筛选黄硝创伤方抑菌有效药味。结果:黄硝创伤方拆方对金黄色葡萄球菌最佳抑菌组合为A1B1C1D1E2,预期药效24.8 mm;对肠球菌最佳抑菌组合为A1B1C2D2E2,预期药效17.3 mm。验证试验证明该组合对金黄色葡萄球菌和临床耐药肠球菌的抑菌圈直径分别为(19.89±1.11)mm和(15.58±1.63)mm。结论:黄硝创伤方对金黄色葡萄球菌和临床肠球菌均有较好的抑制作用,根据各药味对2种实验菌D值的贡献,确定黄硝创伤方的优化组合为五倍子和大黄。五倍子与大黄配伍,对金黄色葡萄球菌有协同抑制作用,而对肠球菌无协同作用。Objective:To screen the main antibacterial ingredients in trauma prescription of Huangxiao(HX)and optimize the best compatibility.Methods:Using the in vitro antibacterial effect as an indicator,orthogonal t-value method was used to prepare and evaluate 12 groups of decoctions according to the orthogonal t-value table.The antibacterial experiment was carried out by drug-sensitive paper disk method.Through investigating whether the inhibition zone was present and the size of its diameter,the antibacterial effects of different prescription group on Staphylococcus aureus standard strain and clinical drug-resistant Enterococcus were determined,and the effective antibacterial herbal medicines of HX were screened.Results:The best antibacterial combination of HX on Staphylococcus aureus was A1B1C1D1E2,and the expected inhibitory zone diameter for evaluating efficacy was 24.8 mm;the best antibacterial combination for Enterococcus was A1B1C2D2E2,and the expected inhibitory zone diameter was 17.3 mm.The validation test proveded that the inhibition zone diameters of the combination against Staphylococcus aureus and clinically resistant Enterococcus were(19.89±1.11)mm and(15.58±1.63)mm,respectively.Conclusion:HX had the good inhibitory effect on Staphylococcus aureus and clinical Enterococcus.According to the contribution of each drug on the D values of the two experimental bacterial strains,the optimal combination of HX was determined to be gallnut and rhubarb.There was a synergistic inhibitory effect on Staphylococcus aureus when combination gallnut and rhubarb,while no synergistic effect on Enterococcus was found.
